Abstract

MSG merupakan hasil purifikasi glutamat sintesis yang dapat menjadi makronutrient penyusun komponen seluler. Tujuan dalam penelitian ini adalah untuk mengetahui pengaruh pemberian MSG terhadap pertumbuhan isolat bakteri Bacillus PL01 dan Pseudomonas PL01 pada MSM. Sebagai kontrol digunakan medium Lactose Broth (LB). Pertumbuhan diukur setiap 1 jam sekali selama 48 jam. Parameter yang diukur adalah Optical Density (OD) λ 600nm, jumlah sel bakteri, serta pengukuran diameter sel. Data dianalisis secara deskriptif kuantitatif dengan Analysis of Variance (ANOVA). Pada Bacillus PL01 pertumbuhan tertinggi terjadi setelah 16 jam masa inkubasi pada konsentrasi 0,5 gr/L MSG dengan nilai serapan OD 600nm yang setara dengan 3.93E+06 Sel/mL serta ukuran sel mencapai 1,8μm x 1,2μm, dimana pertumbuhannya melebihi pertumbuhan di medium LB. Namun terjadi penurunan pertumbuhan seiring dengan peningkatan konsentrasi MSG. Pada Pseudomonas PL01 pertumbuhan tertinggi setelah 16 jam masa inkubasi terjadi pada konsentrasi 2 gr/L MSG dengan nilai serapan OD 600nm yang setara dengan 9.97E+05 Sel/mL serta ukuran sel mencapai 2,4μm x 1,1μm. Pertumbuhan Pseudomonas PL01 masih berada dibawah LB. Berbanding terbalik dengan Bacillus PL01, pada Pseudomonas v PL01 semakin tinggi konsentrasi MSG, maka pertumbuhannya semakin tinggi pula. ========================================================= MSG is a synthetic purified glutamate that can be used as a nitrogen source for microorganisms growth. The purpose of this research was to determine the effect of MSG on Bacillus PL01 and Pseudomonas PL01 growth in a Mineral Salt Medium. As a control medium Lactose Broth (LB) was used. The bacterial growth was measured every 1 hour for 48 hours spectrophotometrically (OD 600nm), cells number using was counted and cell shape was measured. Data were analyzed statistically using a Quantitative with Analysis of Variance (ANOVA). In 0,5 gr/L MSG containing MSM, Bacillus PL01 growth a highest speed after 16 hours it reached OD 600nm that equals to 3.93E+06 Cell/mL, and the cell size was around 1,8μm x 1,2μm. This growth was even higher compared with it in LB. While Pseudomonas PL01 reached the highest growth in 2 g/L MSG containing MSM with OD 600nm that equals to 9.97E+05 Cell/mL with the cell size about 2,4μm x 1,1μm, but Pseudomonas PL01 was more prefer to grow in LB at which its growth was the best. The growth was increase over the MSG concentration, even they were still lower than it in LB.
