Abstract

Di Indonesia, kebutuhan akan Zea mays tiap tahunnya
meningkat. Hal ini berbanding terbalik dengan sedikitnya lahan
pertanian yang digunakan. Solusinya yakni memanfaatkan
lahan marginal dengan kandungan garam tinggi. Tujuannya
adalah untuk menguji pengaruh tingkat ketahanan varietas
Talango dan Manding terhadap salinitas serta melihat adanya
keragaman genetik galur hasil seleksi in vitro.
Pada penelitian ini, untuk mendapatkan varietas Z. mays
yang toleran terhadap salinitas dilakukan dengan
menggunakan teknik in vitro. Pertama dilakukan induksi kalus
(MS0+4ppm 2,4-D), kemudian kalus disubkultur pada medium
seleksi (MS0+4ppm 2,4-D+konsentrasi NaCl (0, 2500, 5000,
dan 7500 ppm)). Selanjutnya dilakukan analisis keragaman
genetik dengan RAPD.
Parameter ketahanan kalus pada perlakuan dapat dilihat
dari perubahan morfologi warna kalus, persentase kalus hidup,
pertambahan berat kalus yang semakin menurun. Analisis
RAPD menunjukkan hasil bahwa terjadi polimorfisme pada
kedua varietas dengan primer OPA 13, OPB 07, OPC 02, OPK
20, dan OPU 19. Hal ini membuktikan adanya perbedaan
genetik antara kalus kontrol dan toleran.

In Indonesia, needs for Zea mays annually increased. It is
inversely proportional to fewer agricultural land that can be
used. The solution that is utilizing marginal land with high salt
content. The aim of this study are to test the effect of resistance
level Talango and Manding varieties to salinity and to know the
genetic diversity of in vitro selection yield strains.
In this study, to obtain salinity tolerant Z. mays varieties,
used in vitro techniques. First, callus was induced (MS0+4ppm
2,4-D), then subcultured on selection medium (MS0+4ppm 2,4-
D+NaCl concentration (0, 2500, 5000, and 7500 ppm)). Then
analysis of genetic diversity used RAPD.
Callus resistance parameter on treatment could be seen
by the callus morphological change of color, alive callus
percentage, callus weight gain decreasing with increasing
salinity concentration. Analysis RAPD showed that
polymorphism occurs in both varieties with primer OPA 13,
OPB 07, OPC 02, OPK 20, and OPU 19. This proves the
existence of genetic differences between control and tolerant
callus.
