Abstract

Enzim lipolitik merupakan enzim yang memiliki peranan
penting dalam bioteknologi modern. Enzim ini mampu
menghidrolisis ikatan ester dan memecah trigliserida menjadi
asam lemak bebas dan gliserol. Enzim lipolitik banyak dihasilkan
dari mikroorganisme, salah satunya adalah kapang Aspergillus
niger. Tujuan dari penelitian ini adalah untuk mengetahui daya
hidup dan daya kerja kapang Lipolitik Aspergillus niger pada
medium minyak goreng bekas dan air pencucian ikan pada
periode tertentu.
Metode yang digunakan dalam penelitian ini meliputi uji
viabilitas, pengukuran biomassa, pengukuran aktivitas enzim,
serta pengukuran jumlah degradasi lipid. Masa inkubasi yang
dilakukan pada penelitian ini selama dua bulan dengan
perlakuan setiap dua minggu sekali mulai dari minggu ke-1
hingga minggu ke-9 dengan menggunakan limbah pencucian ikan
dan minyak goreng bekas. Hasil penelitian menunjukkan bahwa
Aspergilllus niger mampu mempertahankan viabilitasnya pada
kedua medium selama 9 minggu. Aktivitas lipolitik dan degradasi
lipid tertinggi yaitu sebesar 0,12U/mL dan 91%pada medium
limbah pencucian ikan.

Lipolytic enzyme is an enzyme that has an important role
in biotechnology. Lipolytic enzyme can hydrolyze the ester bonds
and break down triglycerides into free fatty acids and glycerol.
Many lipolytic enzymes produced by microorganisms, one of
which is Aspergillus niger. The purpose of this study was to
determine the viability and ability of lipolytic fungus Aspergillus
niger in used-cooking oil and fish-washing waste water medium
in certain periods.
The method used in this study include the viability test,
measurement of biomass, enzyme activity measurement, and
measurement of lipid degradation amount. The incubation period
on this study is two months. The treatment was done in every two
weeks, from week 1 to week 9, using fish-washing waste and
used-cooking oil. The results of this study is Aspergilllus niger
able to maintain viability in both medium for 9 weeks. The
highest lipolytic activity and lipid degradation is 0,12U/mL and
91% on fish-washing waste water medium.
