Abstract

Plastik merupakan polimer sintetis yang terbuat dari unsurunsur
bahan bakar fosil. Polietilen merupakan salah satu jenis
plastik yang banyak digunakan sebagai bahan dasar pembuatan
kantong plastik belanja. Plastik akan menyebabkan masalah bagi
lingkungan karena sifat plastik yang susah terdegradasi.
Degradasi plastik di lingkungan dapat terjadi secara biologis
menggunakan mikroorganisme. Penelitian ini bertujuan untuk
mengetahui potensi mikroorganimse air sampah dan inokulum
campur koleksi laboratorium, serta yeast M 2.3 dalam
mendegradasi plastik.
Metode biodegradasi plastik yang digunakan adalah kolom
Winogradsky. Penelitian ini menggunakan botol berukuran 600
ml yang berisi 300 gram pasir steril, medium MSM 350ml,
inokulum air sampah (S) , inokulum campur (X) serta yeast M 2.3
(Y), dan plastik uji hitam, putih serta transparan dengan 3 kali
pengulangan destruktif. Perhitungan degradasi plastik dilakukan
setiap 3 minggu sekali selama 3 bulan dengan menggunakan
Rancanagan Acak Lengkap (RAL). Analisis data menggunakan
ANOVA-One way dengan uji lanjutan Tukey’s multiple range.
Hasil yang didapat adalah inokulum X, S dan Y memiliki
kemampuan degradasi plastik, namun persentase degradasi
ketiga inokulum tersebut tidak berbeda nyata terhadap plastik
hitam, putih maupun transparan. Pada plastik hitam degradasi
optimal terjadi setelah 6 minggu masa inkubasi oleh inokulum S
mencapai 4,9%. Plastik putih degradasi optimal terjadi setelah 9

minggu masa inkubasi oleh inokulum X dengan nilai 3,6% dan
pada plastik transparan terjadi pada inokulum S setelah 9
minggu masa inkubasi dengan persentase degradasi 4%. Koloni
bakteri yang diduga potensial adalah XH6, XP1, XP9, XT1, XT2,
SH1, SP1, ST1, ST2,dan ST7, serta koloni yeast adalah XP1,
XT1, SH1 dan SP1.

Plastic is a synthetic polymer made from elements of fossil
fuels. Polyethylene is a type of plastic which is widely used as
basis for making shopping plastic bags. Plastics would cause
problems for the environment because of their characteristic that
hardly degradable. Plastic degradation in the environment could
occur biologically using microorganisms. This study aims to
determine the potential of waste water microorganisms and the
mixed inoculum from laboratory collection, also yeast M 2.3 in
the plastic degradation.
Plastic biodegradation method which used is Winogradsky
column. This research was using 600 ml size bottles containing
300 grams of sterile sand, 350ml MSM medium, inoculum waste
water (S), mixed inoculum (X) and yeast 2.3 M (Y), black plastic
test, white plastic test, and transparent plastic test with 3
repetitions destructive. Plastic degradation calculations performed
once every 3 weeks for 3 months using the Completely
Randomized Design (CRD). Analysis of data was using One way
ANOVA with Tukey test following up.
The results obtained are inoculum X, S and Y have the
capability to degrade plastic, but the percentage of degradation of
the three inoculum were not significantly different with black
plastic, white plastic or transparent plastic. Black plastic is the best
degradation ability about 4,9% by inoculum S, white plastic is the best
degradation ability about 3,6% by inoculum X and transparent plastic is

the best degradation ability about 4% by inoculum S. Bacterial
colonies which suspected potentially are XH6, XP1, XP9, XT1,
XT2, SH1, SP1, ST1, ST2, and ST7, as well as yeast colonies are
XT1, XP1, SH1 and SP1.
