Abstract

Kromium (Cr) tergolong logam berat yang dapat masuk ke lingkungan melalui limbah industri sehingga mencemari lingkungan. Bakteri resisten Cr mampu mengubah Cr(VI) menjadi Cr(III). Penelitian ini bertujuan untuk mengetahui resistensi dan potensi bioremoval kromium oleh Bacillus koleksi Laboratorium Mikrobiologi dan Bioteknologi Jurusan Biologi ITS Surabaya. Penelitian diawali dengan uji resistensi 6 isolat Bacillus untuk mengetahui kemampuan tumbuh isolat pada medium yang mengandung Cr. Dilakukan range finding test untuk mendapatkan konsentrasi logam berat sebagai perlakuan bioremoval dan menentukan 3 isolat terpilih. Kepadatan sel diukur dengan OD menggunakan spektrofotometer UV-Vis dan divisualisasikan dengan kurva pertumbuhan. Bioremoval Cr diukur dengan metode Atomic Absorption Spectroscophy selanjutnya dilakukan uji viabilitas dengan metode pour plate. Hasil penelitian menunjukkan isolat Bacillus resisten dan viabel terhadap logam Cr pada konsentrasi ≤ 150 mg/L. Pola pertumbuhan isolat Bacillus yang tercekam logam Cr relatif lebih rendah dibandingkan yang tidak tercekam Cr. Efisiensi bioremoval Cr yang paling tinggi adalah Bacillus DA11 pada konsentrasi perlakuan 38.8 mg/L, sebesar 76.8%, ================================================================================================= Chromium (Cr) is one of heavy metal that encountered environment from industrial waste so it can contaminate the environment. Chromium resistance bacteria can transform Cr(VI) to Cr(III). The aim of this research is to find out chromium resistance and bioremoval potential of Microbiology and Biotechnology Laboratory’s collection Bacillus spp. This research was started with resistance assay of 6 isolates Bacillus to determine growth ability of isolates on medium contained Cr. Range finding test were done to determine heavy metal concentration that used in research and determine 3 isolates that more resistance than others. Cell density were measured by OD using spectrophotometer UV-Vis and visualized by growt curve. Chromium bioremoval was measured by Atomic Absorption Spectroscophy method. Viability assay was done by pour plate method. The result showed that Bacillus resistant and viable on media contained chromium at concentration ≤ 150 mg/L. Growth pattern of Bacillus grown in media contained Cr was relative lower than control. DA11 got highest bioremoval efficiency percentage in the amount of 76.8% at concentration 38.8 mg/L
