Abstract

Protease (EC 3.4) merupakan salah satu kelas enzim yang
penting dalam aplikasi bioteknologi dan industri dan menempati
sekitar 60% dari total penjualan enzim. Enzim protease banyak
dihasilkan dari mikroorganisme, salah satunya adalah marine
yeast. Produksi enzim protease dipengaruhi oleh beberapa faktor
seperti masa inkubasi, suhu dan pH. Tujuan dari penelitian ini
adalah untuk mengetahui masa inkubasi serta suhu dan pH yang
optimal untuk produksi enzim protease dari Candida G3.2.
Optimasi produksi protease diawali dengan optimasi masa
inkubasi dilanjutkan dengan optimasi suhu dan pH. Variasi masa
inkubasi yang digunakan adalah 0, 24, 48, 72, 96, 120, 144 dan
168 jam. Variasi suhu yang digunakan adalah suhu ruang, 45°C
dan 55°C. Sedangkan variasi pH yang digunakan adalah 4,5, 7
dan 9. Pengujian optimasi produksi protease didasarkan pada
aktivitas enzim yang dihasilkan dengan tirosin sebagai produk
yang diukur. Desain penelitian untuk perlakuan suhu dan pH
menggunakan rancangan faktorial. Data aktivitas enzim untuk
masa inkubasi dianalisis secara deskriptif kuantitatif. Sedangkan
data aktivitas enzim untuk suhu dan pH dianalisis secara statistik
menggunakan Anova Two-Way. Hasil penelitian menunjukkan
bahwa Candida G3.2 mampu memproduksi enzim protease secara
optimal pada masa inkubasi 72 jam dengan nilai aktivitas enzim
sebesar 321,5 U/mL serta pada suhu 55°C dan pH 9 dengan nilai
aktivitas enzim 135,25 U/mL.

Protease (EC 3.4) is one of an enzyme classes which
important in biotechnology and industrial applications and
occupying about 60% of the total sales of enzyme. Protease could
be produced by microorganism, one of those is marine yeast.
Protease production is highly influenced by several factors, such
as incubation time, temperature and pH. The aim of this study is
to determine the incubation time and temperature and pH which is
optimal for the production of protease from Candida G3.2.
Optimization of protease enzyme production started with
optimization of incubation period and followed by optimization
of temperature and pH. Variations of the incubation period used
are 0, 24, 48, 72, 96, 120, 144 dan 168 hours. Variations of
temperature used are room temperature, 45°C and 55°C. While
the pH variations used are 4.5, 7 and 9. The optimization protease
production assay is based on the enzyme activity with tyrosine as
measured product. The research design for temperature and pH
used is factorial design. The enzyme activity data for incubation
time were analyzed descriptively quantitative. Meanwhile, the
enzyme activity data for temperature and pH were analyzed
statistically using ANOVA Two-Way. The results showed that
Candida G3.2 is capable of producing protease optimally on 72
hour of incubation period with the value of enzyme activity is
321.5 U/mL and at 55°C and pH 9 with the value of enzyme
activity is 135,25 U/mL.
