Abstract

Jagung (Zea mays L.) merupakan komoditas pangan strategis dan bernilai ekonomis tinggi serta mempunyai peluang untuk dikembangkan karena kedudukannya sebagai sumber utama karbohidrat dan protein setelah beras. Permintaan jagung yang terus meningkat dari tahun ke tahun tidak linear dengan peningkatan produksi tanaman jagung dikarenakan adanya alih fungsi lahan sehingga, diperlukan area baru yaitu dengan memanfaatkan lahan marginal seperti lahan salin di pesisir. Tujuan dari penelitian ini adalah untuk mengetahui tingkat ketahanan dan pengaruh cekaman salinitas pada jagung varietas Bluto dan Manding serta keragaman genetik galur hasil seleksi in vitro. Pada penelitian ini, untuk mendapatkan varietas Z. mays yang toleran terhadap salinitas dilakukan dengan menggunakan teknik in vitro. Pertama dilakukan induksi kalus (MS0 + 2,4-D 3 ppm), kemudian kalus disubkultur pada medium seleksi (MS0 + 2,4-D 3 ppm + NaCl (0, 2500, 5000, dan 7500 ppm)). Selanjutnya dilakukan analisis RAPD untuk melihat keragaman genetik.

Maize (Zea mays L.) is a strategic food commodities and has a high economic value as well as have the opportunity to be developed due to its fuction as the main source of carbohydrates and protein after rice. The demand of maize continues to increase years to come and this condition is not linear with the production level due to land conversion. So that needed a new area for the production of maize that is by utilizing marginal land as saline coastal land. The aims of this study is to investigate the level of resistance and the effect of salinity on Maize varieties i.e Bluto and Manding and also the genetic diversity of somaclon resulted from in vitro selection. In this study, in vitro selection will be applied to obtain high salt tolerance varieties of Z. mays. First, callus induction will be conducted by using medium with composition MS0 + 2,4- D 3ppm, then subcultured onto selection medium with the composition MS0 + 2,4-D 3 ppm + NaCl (0, 2500, 5000, and 7500 ppm). Furthermore, RAPD analysis is used to see genetic diversity
