Abstract

Gelatin merupakan biopolimer produk denaturasi kolagen yang banyak dimanfaatkan dalam berbagai bidang. Sifat gelatin dipengaruhi oleh crosslink pada kolagen, dimana perbedaan crosslink kolagen dipengaruhi oleh ukuran atau usia hewan. Penelitian ini bertujuan untuk mengetahui pengaruh ukuran tubuh terhadap kemurnian dan gugus fungsi gelatin kulit ikan patin (P. hypophthalmus). Kulit ikan patin dengan ukuran meliputi ikan kecil (total ukuran panjang tubuh 38,9 ± 0,5 cm) dan ikan besar (total ukuran panjang tubuh 48,6 ± 1,9 cm) diekstraksi dengan pretreatment asam dan basa untuk memperoleh kolagen, kemudian didenaturasi menjadi gelatin. Analisis kemurnian gelatin dilakukan dengan menggunakan RP-HPLC menghasilkan nilai kemurnian 34,36-86,24% untuk gelatin kulit ikan patin ukuran kecil dan 67,57-85,59% untuk gelatin kulit ikan patin ukuran besar. Analisis spektrum FTIR gelatin kulit ikan patin ukuran kecil dan besar menunjukkan bilangan gelombang yang hampir sama pada daerah serapan amida A, B, I, II, dan III. Pada kedua gelatin kulit ikan patin ukuran kecil dan besar terdapat gugus fungsi O-H, C-H, N-H, C=O, C=N, C=C, dan N=O ======================================================================================================================== Gelatin is a biopolymer of collagen denaturation products that is widely used in various industry. The nature of gelatin is influenced by crosslinks on collagen, differences in collagen crosslinks are influenced by the size or age of the animal. The aim of this study were to determine the effect of body size on the purity and functional groups of striped catfish skin gelatin (P. hypophthalmus). Striped catfish skins of different sizes include small fish (total body length 38.9 ± 0.5 cm) and large fish (total body length 48.6 ± 1.9 cm) extracted with acid and base pretreatment to obtain collagen, then denatured to gelatin. Purity of gelatin were analysis by RP-HPLC and result purity values 34.36-86.24% for small size striped catfish skin gelatin and 67.57-85.59% for large striped catfish skin gelatin. FTIR spectra analysis of small and large striped catfish skin gelatin result wavenumbers that are almost same in amide A, B, I, II, and III region. In both striped catfish skin gelatin small and large sizes there are O-H, C-H, N-H, C = O, C = N, C = C, and N = O functional groups
