Abstract
Merkuri adalah logam berat yang sangat tosik dan
berbahaya. Beberapa bakteri dapat hidup di habitat yang
tercemar merkuri disebut bakteri resisten merkuri, satu
diantaranya adalah Azotobacter.
Tujuan penelitian adalah mendapatkan isolat unggul
Azotobacter dari lahan Eco Urban Farming ITS yang resisten
terhadap HgCl2 dan mengetahui kemampuan aktivitas enzim
merkuri reduktase sebagai bioreduktor Hg
2+ 0menjadi Hg. Isolasi dilakukan menggunakan media selektif Azotobacter agar.
Pengujian resistensi merkuri dilakukan dengan metode goresan
agar miring yang megandung HgCl2. Aktivitas enzim merkuri
reduktase dianalisis dengan metode MRA dan spektofotometri
pada panjang gelombang 340 nm.
Hasil isolasi mendapakan 10 isolat Azotobacter resisten
HgCl2, isolat A5 dan A9 mempunyai resistensi 10 mg/L HgCl2
dengan pertumbuhan yang optimum dan mempunyai aktivitas
merkuri reduktase relatif tinggi dengan daya reduksi ± 3 mg/L dan efisiensi reduksi >70% pada konsentrasi 5 mg/L HgCl2

Mercury is a heavy metal that its dangerous toxic. Some
bacteria can living at contaminated mercury areas, called mercury
resistant bacteria which one of them Azotobacter.
The purpose of this researched to get Azotobacter from
Eco Urban Farming ITS land which resistant mercury HgCl2 and
determine activity of mercury reductase enzyme for reduction

. Methods isolation used selective Azotobacter agar,
mercury resistant with added HgCl2 different concentration, and
activity of mercury reductase enzyme analyzed MRA method and
spechtophotometry at 340 nm.
The result of isolation got 10 isolate Azotobacter what
resistant mercury HgCl2, isolate A5 and A9 resistant 10 mg/L.
with optimum growth and they have activity of mercury reductase
enzyme more higher with reduction abilities ± 3 mg/L and
efficiency reduction Hg
2+
