Abstract
Liofilisasi adalah teknik konservasi dengan proses sublimasi cepat dalam kondisi vakum yang dapat diterapkan untuk produksi biofertilizer komersial. Liofilisasi menghasilkan produk bakteri kering yang stabil secara struktur, viabilitas bakteri yang tinggi pada penyimpanan yang lama dan mudah ditransportasikan. Salah satu tahapan penting dalam liofilisasi adalah kultur bakteri dan agen lioprotektan yang menjaga viabilitas sel selama penyimpanan. Tujuan studi literatur ini adalah untuk mencari medium pertumbuhan alternatif dan agen lioprotektan terbaik untuk Pseudomonas sp., Bacillus sp., Rhizobium sp., Azotobacter sp., dan Azospirillum sp. sebagai agen potensial biofertilizer. Data dari jurnal, prosiding dan laporan penelitian, disusun dan dianalisis secara deskriptif. Hasil studi literatur menunjukkan bahwa lentil hitam (Vigna mungo), kacang hijau (Vigna radiata), splitpea yellow (Pisum sativum), kacang tunggak (Vigna unguiculata), ubi kuning (Ipomea batatas) dan ubi ungu (Dioscorea alata) mampu menjadi medium alternatif sumber nutrisi. Jumlah pertumbuhan bakteri di medium tersebut lebih banyak daripada medium paten nutrient broth dan nutrient agar. Sedangkan agen lioprotektan terbaik adalah sukrosa 5-10% dan skim milk 10-20% untuk sel tidak terimobilisasi atau skim milk 6-10% untuk sel terimobilisasi.

Lyophilization is a conservation technique with rapid sublimation process under vacuum condition which can be applied for commercial biofertilizers production. Lyophilization produces dry bacterial product which is stable in structure, has high bacterial viability after a long storage period and easy to be transported. One of the important steps in lyophilization technique are culturing bacteria on growth medium and lyoprotectant agent to keep cells - viable during long storage. The aim of this literature study was to find alternative growth media and the best lyoprotectant agents for Pseudomonas sp., Bacillus sp., Rhizobium sp., Azotobacter sp., and Azospirillum sp. as biofertilizer potential agent. Data from journal publications, proceedings and research reports were compiled and descriptively analyzed. The results showed that black gram (Vigna mungo), green gram (Vigna radiata), splitpea yellow (Pisum sativum), cowpea (Vigna unguiculata), yellow sweet potato (Ipomea batatas) and purple sweet potato (Dioscorea alata) were able to provide bacteria need nutrients. The number of bacterial growth on those medium were more than they were on patent nutrient broth and nutrient agar. While, the best lyoprotectant agent were sucrose 5-10% and skim milk 10-20% for non-immobilized cells or skim milk 6-10% for immobilized cells.
