Abstract
Perkembangan Industri tekstil dan produk tekstil (TPT) di Indonesia yang terus meningkat mengakibatkan limbah industri tekstil juga ikut meningkat. Salah satu jenis limbah hasil dari industri tekstil adalah pewarna Reactive Black 5 (RB5). Keberadaan RB5 dalam limbah industri dapat mencemari lingkungan perairan dan memberikan dampak buruk bagi kesehatan maupun ekosistem makhluk hidup karena sifatnya yang karsinogenik dan sulit terdegradasi secara alami. Proses degradasi yang efektif diperlukan untuk mendegradasi pewarna tekstil RB5 agar tidak berbahaya bagi lingkungan. Pada penelitian ini dilakukan biodegradasi pewarna tekstil RB5 oleh bakteri B. subtilis pada media cair LB broth. Pengaruh konsentrasi, waktu inkubasi, suhu inkubasi, pH, dan perlakuan digoyang dan didiamkan diuji untuk mengetahui kondisi optimum B. subtilis dalam mendegradasi RB5. Kondisi optimum degradasi RB5 oleh bakteri B. subtilis yaitu menggunakan konsentrasi pewarna RB5 50 mg/L, waktu inkubasi 48 jam, suhu 35 ℃, pH 7, dan diinkubasi dengan didiamkan. B. subtilis mampu mendegradasi RB5 hingga 89,76%. Hasil menunjukkan bahwa B. subtilis dengan kondisi optimumnya memiliki kemampuan dalam mendegradasi pewarna RB5.

The development of textile and textile products industries in Indonesia resulted in increasing textile industry waste. One type of waste from the textile industry is Reactive Black 5 (RB5) dye. The presence of RB5 in industrial waste causes some negative effects on aquatic environment and affect health and ecosystem of living things due to its carcinogenic and difficult to degrade naturally. Thus, the degradation process is required to degrade RB5 textile dye into unharmful compounds. In this study, biodegradation of textile dye RB5 by B. subtilis bacteria using LB broth media was investigated. The effect of concentration, incubation time, temperature, pH, and static and shaker conditions were investigated to determine the optimum condition of B. subtilis in degrading RB5. The optimum conditions of RB5 degradation by B. subtilis were 50 mg/L dye concentration, 48 hours incubation time, temperature 35 °C, pH 7, and incubated in static condition. B. subtilis was able to degrade 89.76% of RB5. The results showed that B. subtilis with its optimum conditions had the ability to degrade RB5 dyes.
