Abstract
Identifikasi bakteri Streptococcus pyogenes dan Moraxella catarrhalis telah berhasil dilakukan. Dalam penelitian ini dilakukan variasi penumbuhan bakteri Streptococcus pyogenes dan Moraxella catarrhalis yang masing-masing ditumbuhkan pada tiga cawan agar yang berbeda dan dibuat rangkaian pengenceran dalam pelarut Phosphate Buffered Saline (PBS) pH 7,4. Sampel uji penelitian ini berasal dari hasil menumbuhkan bakteri pada cawan, dengan populasi yang hampir sama yakni untuk Streptococcus pyogenes pada cawan 1 adalah 2,21 × 107 CFU/ml, cawan 2 sebanyak 1,95 × 107 CFU/ml, dan cawan 3 adalah 1,99 × 107 CFU/ml. Sedangkan untuk Moraxella catarrhalis menggunakan cawan 1 dengan jumlah koloni sebanyak 2,18 × 107 CFU/ml, cawan 2 adalah 2,24 × 107 CFU/ml, dan cawan 3 sebesar 2,06 × 107 CFU/ml. Selanjutnya sampel uji diukur menggunakan spektrofotometer fluoresensi dan diperoleh panjang gelombang pada eksitasi dan emisi maksimumnya untuk membedakan jenis antar sampel uji. Analisis sampel dibantu dengan metode Principal Component Analysis serta uji signifikansi ANOVA dan LSD. Berdasarkan hasil PCA serta uji signifikansi ANOVA dan LSD untuk panjang gelombang pada eksitasi dan emisi maksimum, menunjukkan bahwa spektrofotometer fluoresensi dapat membedakan kedua jenis bakteri dalam pelarut PBS pH 7,4.

The identification of Streptococcus pyogenes and Moraxella catarrhalis bacteria has been successfully carried out. In this study, variations in the growth of Streptococcus pyogenes and Moraxella catarrhalis bacteria were carried out, each of which was grown in three different agar plates and made a series of dilutions in Phosphate Buffered Saline (PBS) solvent pH 7.4. The test samples of this study were derived from the results of growing bacteria in plates, with almost the same population, for Streptococcus pyogenes, the first plate was 2.21 × 107 CFU/ml, the second plate was 1.95 × 107 CFU/ml, and the third plate was 1.99 × 107 CFU/ml. Meanwhile, for Moraxella catarrhalis first plate had a colony count of 2.18 × 107 CFU/ml, the second plate was 2.24 × 107 CFU/ml, and the third plate was 2.06 × 107 CFU/ml. Furthermore, the test samples were measured using a fluorescence spectrophotometer and obtained the maximum excitation, and maximum emission wavelengths were to distinguish the types between test samples. The sample analysis was assisted by the Principal Component Analysis method and the ANOVA and LSD significance tests. Based on PCA results and ANOVA and LSD significance tests for wavelengths at maximum excitation and emission, it was shown that the fluorescence spectrophotometer could distinguish the two types of bacteria in PBS solvent pH 7.4.
