Abstract

Trisindolina 1 diketahui memiliki potensi sebagai antikanker. Perlu adanya pengujian aktivitas Trisindolina 1 terhadap Cancer Stem Cell (CSC) terutama dengan parameter Reactive Oxygen Species (ROS). Penelitian ini bertujuan untuk menganalisis aktivitas Trisindolina 1 terhadap tingkat sitotoksisitas dan kadar ROS pada CSC. Analisis sitotoksisitas menggunakan metode MTT assay. Adapun analisis kadar ROS menggunakan pewarnaan DCFH2-DA yang dievaluasi dengan flowcytometer. Berdasarkan hasil uji sitotoksisitas, diperoleh nilai IC50 dari Trisindolina 1 sebesar 57,72 μM sehingga tergolong senyawa dengan aktivitas sedang, dan Doxorubicin sebesar 13,55 μM sehingga tergolong senyawa dengan aktivitas baik. Berdasarkan hasil pengukuran kadar ROS, dapat disimpulkan bahwa mekanisme kerja Trisindolina 1 adalah dengan cara menurunkan kadar ROS intraseluler sehingga dapat menghambat pembentukan CSC.

Trisindolina 1 is known to have potential as an anticancer. It is necessary to test the activity of Trisindolina 1 against Cancer Stem Cells (CSC), especially with Reactive Oxygen Species (ROS) parameters. This study aimed to analyze the activity of Trisindolina 1 on the level of cytotoxicity and levels of ROS in CSC. Cytotoxicity analysis using the MTT assay method. As for the analysis of ROS levels using DCFH2-DA staining which was evaluated with a flowcytometer. Based on the results of the cytotoxicity test, the IC50 value of Trisindolina 1 is 57.72 μM so it is classified as a compound with moderate activity, and Doxorubicin is 13.55 μM so it is classified as a compound with good activity. Based on the measurement results of ROS levels, it is certain that the mechanism of action of Trisindolina 1 is to reduce intracellular ROS levels so that it can inhibit the formation of CSC.
