Abstract
Skeletonema costatum merupakan salah satu kelompok mikroalga yang cenderung meningkatkan akumulasi lipid pada kondisi tercekam. Salah satu kondisi yang menyebabkan peningkatan akumulasi lipid adalah cekaman abiotik. Contoh cekaman abiotik yang meningkatkan akumulasi lipid adalah aerasi dan fotoperiode. Cekaman Aerasi dan fotoperiode akan menyebabkan mikroalga mengubah jalur biosintesis asam lemak menuju pembentukan dan akumulasi lipid netral, terutama dalam bentuk TAGs. Lipid pada mikroalga akan dihasilkan melalui proses biosintesis asam lemak dengan menggunakan ACCase. ACCase dikelompokkan menjadi dua bentuk yakni ACCase heteromerik dan ACCase homomerik.. Gen ACC1 dan ACC2 merupakan kedua protein tipe homomerik yang berperan dalam biosintesis asam lemak. Penelitian ini bertujuan untuk mengetahui
ekpresi gen ACC1 dan ACC2 dibawah kondisi cekaman aerasi dan fotoperiode pada mikroalga S. costatum. Metode yang digunakan adalah pengambilan sampel ekstrak RNA mikroalga S. costatum, desain primer, analisis ekpresi gen dengan q-PCR kemudian dihitung dan analisis data secara deskriptif. Hasil penelitian menunjukkan bahwa Gen ACC1 terekspresi dan mengalami up-regulated pada perlakuan (A2F2): Aerasi 24 jam dan Fotoperiode 24:0 dengan nilai CT: 36,85. Gen ACC2 terekspresi dan mengalami up regulated pada perlakuan (A1F1): Aerasi 12 Jam dan Fotoperiode 12: 12 dengan nilai CT sebesar 36,874. Gen ACC1 pada perlakuan (A1F1): Aerasi 12 Jam dan Fotoperiode 12: 12 dan Gen ACC2 tidak terekspresi pada perlakuan (A2F2): Aerasi 24 jam dan Fotoperiode 24:0 tidak teridentifikasi yang ditunjukkan dengan nilai CT adalah 0. Studi literatur dilakukan untuk mengetahui tren ekspresi kedua gen dibawah cekaman aerasi dan fotoperiode. Gen ACC1 dan ACC2 cenderung mengalami peningkatan ekspresi (upregulated).

Skeletonema costatum is a group of microalgae that tends to increase lipid accumulation in stressed conditions. One of the conditions that cause increased lipid accumulation is abiotic stress. Examples of abiotic stresses that increase lipid accumulation are aeration and photoperiod. Aeration stress and photoperiod will cause microalgae to change the fatty acid biosynthetic pathway towards the formation and accumulation of neutral lipids, especially in the form of TAG. Lipids in microalgae will be produced through a fatty acid biosynthesis process using ACCase. ACCase is grouped into two forms, namely heteromeric ACCase and homomeric. ACC1 and ACC2 genes are both homomeric type proteins that play a role in fatty acid biosynthesis. This study aims to determine the expression of ACC1 and ACC2 genes under conditions of aeration stress and photoperiod in microalgae S. costatum. The method used was sampling S.costatum microalgae RNA extract, primer design, gene expression analysis by q-PCR then calculated and descriptive data analysis. The results showed that the ACC1 gene was expressed and upregulated in the treatment (A2F2): Aeration 24 hours and Photoperiod 24:0 with CT value: 36.85. The ACC2 gene was expressed and upregulated in the treatment (A1F1): Aeration 12 Hours and Photoperiod 12: 12 with a CT value of 36,874. ACC1 gene on treatment (A1F1): Aeration 12 Hours and Photoperiod 12:12 and ACC2 gene on treatment (A2F2): Aeration 24 hours and Photoperiod 24:0 is not identified as indicated by CT value 0. Literature study was conducted to determine the trend of expression of the ACC1 and ACC2 gene under aeration and photoperiod stress. The ACC1 and ACC2 gene tends to be upregulated.
