Abstract

Begonia medicinalis atau yang dikenal dengan benalu batu adalah salah satu spesies dari genus Begonia yang memiliki aktivitas antioksidan. Ekstrak tanaman ini mengandung metabolit sekunder seperti flavonoid, alkaloid, steroid dan
terpenoid. Pada penelitian ini telah dilakukan ekstraksi menggunakan ultrasonik atau Ultrasound-Assited Extraction (UAE). Hasil penelitian menunjukkan bahwa ekstraksi dengan pelarut air pada daun benalu batu menggunakan metode UAE
memiliki rendemen terbesar pada waktu ekstraksi 15 menit. Hasil LCMS menunjukkan adanya kandungan betain, kolin, diisobutilftalat, (-) epikatekin, kuersetin-3β-D-glukosida, kuersetin, isorhamnetin dan beberapa senyawa lainnya.
Ekstrak tersebut memiliki aktivitas antikanker dengan nilai LC50 (μg/ml) pada HeLa, MDA-MB, dan HT-29 berturut-turut adalah 289.69±0.088, 1753.57±0.096, 154.17±0.459, termasuk antioksidan yang kuat dengan IC50 (µg/ml) terukur 6.401±0.04 dari ekstrak, serta berpotensi sebagai imunomodulator. Potensi tersebut ditunjukkan dengan penurunan ekspresi IL-6 dan total NO pada sel makrofag yang diinduksi oleh LPS E. coli dibandingkan dengan kontrol sel yang tidak diberi perlakuan ekstrak dan peningkatan fungsi fagositosis P. aeroginosa oleh sel makrofag dengan penambahan ektrak dibandingkan dengan kontrol tanpa ekstrak. Hasil penelitian ini menunjukkan bahwa ekstrak 50 µg/ml memberikan hasil
penurunan yang maksimum dengan konsentrasi IL-6 sebesar 146.50±0.68 pg/ml dibandingkan dengan kontrol sel 195.72±0.66 pg/ml dan penurunan konsentrasi total NO sebesar 55.17±2.88 µM dibandingkan kontrol dengan 121.83±2.88 µM,
dan ekstrak 5 µg/ml memberikan peningkatan fungsi fagositosis terbaik dengan persentase kematian bakteri sebesar 66.06±0.002% dibandingkan kontrol dengan 59.55±0.002%.

Begonia medicinalis or known as benalu batu is a species of the genus Begonia that has antioxidant activity. This plant extract contains secondary metabolites such as flavonoids, alkaloids, steroids and terpenoids. In this study, Ultrasound-Assited Extraction (UAE) was carried out. The results showed that the extraction with water as a solvent on benalu batu leaves using the UAE method had the largest yield at an extraction time of 15 minutes. The LCMS results showed the presence of betaine, choline, iisobutylphthalate, (-) epicatechin, quercetin-3β-D�glucoside, quercetin, isorhamnetin and several other compounds. The extract had
anticancer activity with LC50 (μg/ml) values in HeLa, MDA-MB, and HT-29 were 289.69±0.088, 1753.57±0.096, 154.17±0.459 respectively, including strong antioxidants with IC50 (µg/ml) measured 6.401±0.04 from the extract, and has
potential as an immunomodulator. This potential was shown by the decreased expression of IL-6 and total NO in macrophage cells induced by E. coli LPS compared to control cells that were not treated with extract and increased phagocytosis of P. aeroginosa by macrophage cells with the addition of extract compared to control without extract. The results of this study showed that the 50 µg/ml extract gave the maximum reduction in IL-6 concentration of 146.50±0.68 pg/ml compared to control cells 195.72±0.66 pg/ml and a decrease in total NO concentration of 55.17±2.88 M compared to control with 121.83 ± 2.88 M, and 5µg/ml extract gave the best increase in phagocytosis function with the percentage
of bacterial death of 66.06±0.002% compared to control with 59.55±0.002%.
