Abstract

Metilen biru (MB) merupakan salah satu pewarna yang digunakan dalam industri teksil, akan tetapi dapat menimbulkan bahaya bagi lingkungan dan manusia sehingga diperlukan cara untuk menghilangkannya. Penelitian ini mempelajari biodekolorisasi pewarna MB oleh jamur filamen Aspergillus oryzae (AO) yang terimobilisasi dalam matriks SA/PVA/Bentonit. Sistem imobilisasi menggunakan metode entrapment, dengan komposisi matriks SA 2%/PVA 4%/Bentonit 1% (b/v). Matriks SA/PVA/Bentonit disintesis dalam pelarut air pada suhu 100 °C selama 2 jam. Proses optimasi dari kultur jamur dilakukan dengan metode free cell dan imobilisasi, dengan variasi perlakuan jamur hidup dan mati. Biodekolorisasi MB dilakukan pada media air dengan konsentrasi akhir 100 mg/L dalam keadaan statis dan diamati selama 7 hari. Pengujian kinerja biodekolorisasi MB menunjukkan keunggulan beads SA/PVA/Bentonit/AO mati dalam penghilangan MB dengan persen dekolorisasi (%) yang lebih tinggi daripada SA/PVA/Bentonit dan SA/PVA/Bentonit/AO hidup, yaitu 96,22%. Keberhasilan imobilisasi dibuktikan dengan spektra FTIR dan mikrograf SEM yang hampir sama dengan beads SA/PVA/Bentonit yang telah dilaporkan. Beads imobilisasi memiliki stabilitas dan reusability yang baik setelah empat siklus adsorpsi MB. Hasil ini menunjukkan bahwa A. oryzae yang terimobilisasi dalam matriks SA/PVA/Bentonit dapat digunakan sebagai kandidat baru untuk dekolorisasi MB yang efektif.

Methylene blue (MB) is one of the dyes used in the textile industry, but it can pose a danger to the environment and humans, so a method is needed to remove it. This study investigated the biodecolorization of MB by filamentous fungus Aspergillus oryzae (AO) immobilized into SA/PVA/Bentonite matrix. The immobilization system used the entrapment method, with a matrix composition of SA 2%/PVA 4%/Bentonite 1% (w/v). SA/PVA/Bentonite matrix was synthesized in water solvent at 100 °C for 2 hours. The optimization process of fungal culture was carried out by free cell and immobilization methods, with variations of live and dead fungi. MB biodecolorization was carried out in aqueous media with a final concentration of 100 mg/L under static conditions and observed for 7 days. The performance test of MB biodecolorization showed the superiority of dead SA/PVA/Bentonite/AO beads in the removal of MB with a higher decolorization percentage (%) than live SA/PVA/Bentonite and SA/PVA/Bentonite/AO, approximately 96.22%. The success of immobilization was proven by FTIR spectra and SEM micrographs which were almost the same as the reported SA/PVA/Bentonite beads. The immobilized beads had good cycling and reusability after four repeated cycles of MB adsorption. These results indicate that A. oryzae immobilized on SA/PVA/Bentonite matrix can be used as a new candidate for effective MB decolorization.
