Abstract

Kanker Prostat adalah penyakit keganasan sistem urogenital yang merupakan salah satu penyebab terbanyak kematian pada populasi pria. Faktor risiko untuk kanker prostat termasuk peningkatan usia, riwayat keluarga, obesitas, dan faktor makanan. Androgen Receptor pada prostat memediasi transkripsi dalam proliferasi sel�sel prostat dan mengalami mutasi yang mengarah pada peningkatan proliferasi sel-sel prostat. Peningkatan proliferasi sel�sel prostat ditekan dengan Androgen Receptor sebagai sasaran target. Penelitian ini masih terus dikembangkan. Salah satu tanaman herbal yang berpotensi sebagai antikanker yaitu jahe merah (Zingiber officinale var. Rubrum) yang menyintesis senyawa metabolit (6)-gingerol. Penelitian ini bertujuan untuk mengetahui potensi senyawa (6)-gingerol dalam menghambat Protein Androgen Receptor menggunakan metode in silico dan mengetahui jenis asam amino yang berperan pada ikatan antara
senyawa (6)-gingerol dan Protein Androgen Receptor. Simulasi Docking menggunakan software Pyrx dan visualisasi hasil menggunakan software Discovery Studio. Nilai binding affinity senyawa kontrol dengan AR sebesar -9,0 Kcal/mol dengan nilai RMSD sebesar 0 Å sedangkan nilai binding affinity sebesar -7,5 Kcal/mol dengan nilai RMSD sebesar 0 Å. Asam amino yang memiliki interaksi dengan senyawa 6-gingerol antara lain leusin, asam aspartat, glisin, glutamin, metionin, valin, arginin, threonine, isoleusin dan fenilalanin.

Prostate cancer is a malignant disease of the urogenital system which is one of the most common causes of death in the male population. Risk factors for prostate cancer include increasing age, family history, obesity, and dietary factors. Androgen receptors in the prostate mediate transcription in the proliferation of prostate cells and undergo mutations that lead to increased proliferation of prostate cells. The increase in the proliferation of prostate cells is suppressed with the Androgen Receptor as the target. This research is still be developing. One of the herbal plants that have the potential as an anticancer is red ginger (Zingiber officinale var. Rubrum) which synthesizes metabolite compounds (6)-gingerol. This study aims to determine the potential of the compound (6)-gingerol in inhibiting the Protein Androgen Receptor using the in silico method and to determine the type of amino acid that plays a role in the bond between the compound (6)-gingerol and the Androgen Receptor Protein. Docking simulation using Pyrx software and visualization of results using Discovery Studio software. The binding affinity value of the control compound with AR was -9.0 Kcal/mol with an RMSD value of 0 Å while the binding affinity value was -7.5 Kcal/mol with an RMSD value of 0 Å . Amino acids that interact with 6-gingerol compounds include leucine, aspartic acid, glycine, glutamine, methionine, valine, arginine, threonine, isoleucine, and phenylalanine.
