Abstract

Konsentrasi senyawa bioaktif yang ada pada simplisia tumbuhan, dipengaruhi oleh berbagai faktor, salah satunya adalah metode ekstraksi. Keberhasilan metode ekstraksi bergantung terhadap jenis solvent, temperatur dan durasi ekstraksi. Reflux memiliki kelebihan dalam durasi dan efisiensi solvent, sementara metode maserasi unggul untuk senyawa yang bersifat thermolabile. Optimasi terhadap metode ekstraksi yang tepat diperlukan untuk mendapatkan senyawa dengan efek farmakologi dan efikasi yang tinggi. Rutin adalah senyawa flavonoid yang ditemukan pada Morinda citrifolia dan memiliki farmakologi sebagai antidiabetes. Pada diabetes melitus tipe 2, alpha-glucosidase diekspresikan oleh pasien sebanyak 1.5 kali lipat. Hal ini dapat meningkatkan kadar gula postprandial dalam darah. Inhibitor α-glucosidase (AGI) secara klinis, acarbose, miglitol dan voglibose, senyawa hasil isolasi dari mikroba memiliki efek samping berbahaya. Sedangkan natural product dari tanaman memiliki efek samping dan toksisitas yang lebih rendah. Penelitian ini dilakukan untuk mengetahui pengaruh perbedaan metode ekstraksi pada Morinda citrifolia terhadap penghambatan alpha-glucosidase secara in vitro. Hasil uji one-way ANOVA nilai inhibisi alpha-glucosidase menunjukan adanya signifikansi <0.001 (p < 0.05) metode ekstraksi terhadap aktivitas inhibisi enzim. Fraksi dari ekstraksi reflux memiliki nilai inhibisi 4.63 - 13.68%, sedangkan fraksi maserasi berkisaran antara 3.57– 9.27 %. Fraksi chloroform dari reflux (R1) dan fraksi ethyl acetate-methanol-aquades dari maserasi (M3) menunjukan inhibisi yang lebih tinggi dari acarbose (8.37% ± 0.35) secara berurutan yaitu 13.68% ± 0.26 dan 9.27% ± 0.18. Fraksi lainnya menunjukan nilai inhibisi yang lebih rendah dibandingkan acarbose. Senyawa rutin dideteksi dengan UPLC-MS pada fraksi M3.

Several parameters such as nutrients, time and location of harvesting, and extraction methods affect the extraction yield of the bioactive compound in plants. Reflux and maceration are conventional methods for bioactive plant extraction. It depends on solvent, temperature, and time of matrix extraction. Reflux has a shorter time and is solvent efficient in extraction. Meanwhile, maceration allows extraction of the thermolabile compound. To gain maximum yield of compound extraction, methods need to be optimized. Rutin as a plant flavonoid, found in Morinda citrifolia has a therapeutic effect for antidiabetics. In type 2 Diabetes Melitus, alpha-glucosidase was expressed 1.5 fold higher in diagnosed patients. Leading to an elevation of postprandial glucose level. Clinical Alpha-glucosidase inhibition (AGI) such as acarbose, miglitol and voglibose isolated from microbes have acute adverse side effects. Natural products isolated from plants are compared to be safe and less toxic. The aim of this study is to determine different extraction methods of Morinda citrifolia on in vitro assay of alpha-glucosidase inhibition. One-way ANOVA of percentage inhibition shown value <0.001 (p<0.05) defines significantly different extraction methods toward alpha-glucosidase inhibition. Inhibition activity of reflux fractionation ranges from 13.68 - 4.63%, while maceration fraction ranges from 9.27 - 3.57%. The chloroform fraction of reflux (R1) inhibits 13.68% ± 0.26 of enzyme activity. And ethyl acetate-methanol-aquades fraction of maceration (M3) inhibits enzyme activity by 9.27% ± 0.18, which both have higher inhibition than acarbose at the same concentration (8.37%). Other fractions of each method showed lower inhibition compared to acarbose. Rutin was detected in maceration fraction M3 by UPLC-MS.
