Abstract
Paparan sinar UV merupakan penyebab utama hiperpigmentasi pada kulit. Mengkudu merupakan salah satu bahan alam yang dapat digunakan untuk mengatasi hiperpigmentasi karena mengandung flavonoid dan fenol yang dapat menghambat tirosinase. Pada penelitian ini, ekstraksi senyawa flavonoid dan fenol dilakukan dengan dua metode berbeda yaitu maserasi (ekstraksi dingin) dan refluks (ekstraksi panas). Perbedaan metode ekstraksi yang berbeda dapat memberikan kualitas penarikan senyawa bioaktif yang berbeda pula. Oleh karena itu, diperlukan penelitian lebih lanjut terhadap pengaruh metode ekstraksi buah mengkudu terhadap efektivitas penghambatan aktivitas tirosinase secara in vitro. Pada rancangan penelitian ini, metanol digunakan sebagai solvent pada metode ekstraksi maserasi dan refluks. Penguapan dilakukan pada semua sub-sub fraksi untuk digunakan selama uji in vitro inhibisi aktivitas tirosinase. Buffer fosfat pH 6,5 dan DMSO digunakan sebagai pelarut sampel. L-DOPA digunakan sebagai substrat dari tirosinase. Pengujian dilakukan dengan spektrofotometer pada panjang gelombang 495 nm untuk mengukur L-DOPA. Analisis data menggunakan one way analysis of variance (ANOVA) dengan taraf kepercayaan 95% menunjukkan nilai signifikansi <0,001 (p < 0,005). Uji Tukey menunjukkan bahwa rerata inhibisi tertinggi pada sub-sub fraksi ethyl acetate: MeOH : H2O (5 : 2 : 3) dari kelompok refluks (R5) yaitu sebesar 63,31% ± 0,05 dan terendah pada sub-sub fraksi kloroform 100% dari kelompok maserasi (M1) yaitu sebesar 14,99% ± 0,25.

Exposure to UV rays is the main cause of hyperpigmentation of the skin. Noni is one of the natural ingredients that can be used to treat hyperpigmentation because it contains flavonoids and phenols which can inhibit tyrosinase. In this study, the extraction of flavonoids and phenolic compounds was carried out using two different methods, namely maceration (cold extraction) and reflux (hot extraction). Different extraction methods can provide different withdrawal qualities of bioactive compounds. Therefore, further research is needed on the effect of the noni fruit extraction method on the effectiveness of inhibiting tyrosinase activity in vitro. In this research design, methanol is used as a solvent in the maceration and reflux extraction methods. Evaporation was performed on all sub-fractions to be used during the in vitro tyrosinase activity inhibition test. Phosphate buffer pH 6.5 and DMSO were used as sample solvents. L-DOPA is used as a substrate for tyrosinase. Tests were carried out with a spectrophotometer at a wavelength of 495 nm to measure L-DOPA. Data analysis used one way analysis of variance (ANOVA) with a 95% confidence level indicating a significance value of <0.001 (p <0.005). The Tukey test showed that the highest average inhibition was in the ethyl acetate : MeOH : H2O (5 : 2 : 3) sub-fraction of the reflux group (R5) which was 63.31% ± 0.05 and the lowest was in the 100% chloroform from the maceration group (M1) which was 14.99% ± 0.25.
