Abstract
Cancer stem cell (CSC) adalah proporsi kecil dari populasi sel kanker yang memiliki kemampuan seperti stem cell normal. CSC merupakan pioneer untuk sel kanker dimana memiliki kemampuan self-renewal yang dapat menyebabkan CSC berdiferensiasi menjadi bermacam tipe sel kanker . CSC juga mempunyai sifat resisten terhadap terapi konvensional sehingga suatu senyawa yang mempunyai sifat antikanker kuat dibutuhkan untuk membunuh CSC. Trisindolina diketahui mempunyai potensi sebagai antikanker kuat. Trisindolina sendiri telah disintesis menjadi beberapa modifikasi yang memiliki sifat toksik berbeda dengan modifikasi terbarunya adalah Trisindolina 5. Penelitian ini bertujuan untuk mengetahui nilai IC50 Trisindolina 5. Metode yang digunakan untuk mengatahui IC50 adalah uji sitotoksisitas menggunakan MTT assay. Hasil yang diperoleh adalah nilai IC50 senyawa Trisindolina 5 adalah 18,270 μM pada inkubasi 24 jam tergolong senyawa sangat toksik, 17,067 μM pada inkubasi 48 jam tergolong senyawa sangat toksik dan 52,395 μM pada inkubasi 72 jam sehingga tergolong senyawa dengan sitotoksisitas sedang. Sedangkan nilai IC50 doxorubicin adalah 5,277 μM pada inkubasi 24 jam, 2,334 μM pada inkubasi 48 jam, dan 5,324 μM pada inkubasi 72 jam sehingga tergolong senyawa dengan aktivitas sangat toksik

Cancer stem cells (CSC) are a small proportion of the population of cancer cells that have abilities like normal stem cells. CSC is a pioneer for cancer cells which have the ability to self-renewal which can cause CSCs to differentiate into various types of cancer cells. CSCs are also resistant to conventional therapy, so a compound with strong anticancer properties is needed to kill CSCs. Trisindolina is known to have strong anticancer potential. Trisindolina itself has been synthesized into several modifications that have different toxic properties with the newest modification being Trisindolina 5. This study aims to determine the IC50 value of Trisindoline 5. The method used to determine IC50 is cytotoxicity test using MTT assay. The results obtained were the IC50 values of Trisindolina 5 were 18,270 μM at 24 hours incubation classified as very toxic compound, 17,067 μM at 48 hours incubation classified as very toxic compound and 52,395 μM at 72 hours incubation so it is classified as moderate cytotoxicity. Meanwhile, the IC50 values of doxorubicin were 5,277 μM at 24 hours incubation, 6,285 μM at 48 hours incubation, and 5,324 μM at 72 hours incubation so it is classified as a compound with very toxic activity
