Abstract

Agen hayati seperti khamir dapat digunakan untuk menguraikan crude oil menjadi senyawa yang lebih sederhana dan tidak berbahaya. Isolat khamir W.7.3; G.4.1; PL; LD; SL telah diketahui potensinya dalam mendegradasi crude oil, namun identitas isolat tersebut belum diketahui. Tujuan penelitian adalah mendapatkan nama spesies dan mengetahui hubungan kekerabatannya melalui rekonstruksi pohon filogenetik. Penelitian dilakukan dengan identifikasi secara molekuler meliputi ekstraksi DNA menggunakan PrestoTM Mini gDNA Yeast kit, uji kemurnian DNA menggunakan Spektrofotometer Nanodrop, amplifikasi DNA menggunakan OneTaq® 2X dan MyTaqTM HS Red Mix 2X dan marka Internal Transcribed Spacer, Visualisasi amplikon menggunakan Gel Documenter, analisis sekuens DNA menggunakan BLAST (Basic Local Alignment Search Tool-Nucleotide), dan rekontruksi pohon filogenetik menggunakan MEGA 11. Berdasarkan hasil rekonstruksi pohon filogenetik, terdapat 4 cluster yaitu A (isolat SL dan PL), B (isolat LD), C (isolat W7.3), dan D (siolat G4.1) dengan jarak genetik antar isolat 0,000 – 0,209. Kekerabatan terdekat dimiliki oleh isolat SL dan PL, sedangkan kekerabatan terjauh dimiliki oleh isolat G4.1 terhadap semua isolat. Nilai identity isolat terhadap GenBank ditunjukkan sebesar 99,34% - 100%. Isolat-isolat tersebut teridentifikasi sebagai Candida orthopsilosis (isolat W7.3), Exophiala dermatitidis (isolat G4.1), Candida viswanathii (isolat LD), dan Candida tropicalis (isolat PL dan SL)

Biological agents such as yeast can be used to decompose crude oil into simpler and less harmful compounds. Yeast isolates W.7.3; G.4.1; PL; LD; SL have known potential in degrading crude oil, but the identity of these isolates is not yet known. The purpose of the study was to obtain the name of the species and determine its kinship through phylogenetic tree reconstruction. The research was conducted by molecular identification including DNA extraction using PrestoTM Mini gDNA Yeast kit, DNA purity test using Nanodrop Spectrophotometer, DNA amplification using OneTaq® 2X and MyTaqTM HS Red Mix 2X and Internal Transcribed Spacer markers, amplicons visualization using Gel Documenter, DNA sequence analysis using BLAST (Basic Local Alignment Search Tool-Nucleotide), and phylogenetic tree reconstruction using MEGA 11. Based on the results of phylogenetic tree reconstruction, there are 4 clusters, namely A (isolates SL and PL), B (isolate LD), C (isolate W7.3), and D (isolate G4.1) with genetic distance between isolates 0.000 - 0.209. Isolates SL and PL has the closest relationship, while the farthest relationship is isolate G4.1 toward all isolates. The identity value of isolates against GenBank is shown at 99.34% - 100%. The isolates were identified as Candida orthopsilosis (isolate W7.3), Exophiala dermatitidis (isolate G4.1), Candida viswanathii (isolate LD), and Candida tropicalis (isolates PL and SL).
