Abstract

Kompos merupakan hasil degradasi bahan organik secara
aerob maupun anaerob dengan memanfaatkan potensi
mikroorganisme sebagai aktivator kompos. Kualitas aktivator
kompos sangat tergantung dari kecepatan degradasi yang
dimiliki dan lama masa simpannya. Medium pertumbuhan
berpengaruh penting terhadap masa simpan aktivator kompos
karena untuk mengomptimalkan viabilitas sel dalam waktu yang
lama.
Salah satu komponen medium pertumbuhan adalah nitrogen
berupa protein atau senyawa peptida yang dapat memengaruhi
masa simpan (life span) aktivator kompos. Kompleksitas struktur
protein menyebabkan pemanfaatan nitrogen untuk pertumbuhan
juga berbeda, yang salah satunya berpengaruh terhadap masa
simpan aktivator kompos.
Tujuan penelitian ini adalah mengetahui pengaruh beberapa
sumber nitrogen antara lain yeast extract, pepton, susu skim, dan
rumput laut, yang berpotensi untuk meningkatan masa simpan
aktivator kompos.
Konsorsium aktivator kompos berupa Aspergillus niger dan
Rhizobium sp. dengan perbandingan 1:1 dikultur pada medium
variasi sumber nitrogen dengan konsentrasi yang berbeda yaitu
2,5% dan 1,25% dengan 3 kali pengulangan. Jumlah koloni
dihitung dengan metode TPC (Total Plate Agar), Kadar protein
terlarut pada medium kultur dianalisis dengan metode Bradford.
Hasil kultur aktivator kompos diaplikasikan untuk pembuatan
kompos. Data dianalisis secara deskriptif berupa kurva
pertumbuhan koloni, kemiringan kurva pertumbuhan aktivator
kompos,kadar protein terlarut, suhu kompos, dan tabel analisis
rasio C/N. Hasil penelitian didapatkan bahwa medium rumput
laut konsentrasi 1,25% mampu meningkatkan masa simpan
aktivator kompos karena hasil kemiringan kurva pertumbuhan
relatif rendah sehingga diasumsikan jumlah sel yang cenderung
stabil. Sedangkan nilai rasio C/N terbaik adalah aktivator
kompos pada medium rumput laut konsentrasi 1,25% karena nilai
yang didapatkan sebesar 14,15/1. Sehingga medium rumput laut
konsentrasi 1,25% dapat dimanfaatkan untuk meningkatkan masa
simpan aktivator kompos.

Compost is an aerobic or anaerobic organic material
degradation using microorganism as compost activator.
Microorganism has hydrolisis enzim to accelerate composting
process for degrade organic material. Quality of compost
activator depend on organic degradation time and the life span of
compost activator. The life span of compost activator was
influenced of growth medium to optimize cell viability.
Main component of growth medium is protein which contain
nitrogen sources and has role for compost activator’s life span.
Complexity of protein structure causes difference time to utilize
it, thus this mechanism can enhance activator compost life span.
Therefore, enhacing compost activator can be optimized by
varying nitrogen sources, such as pepton, yeast extract, skim
milk, and seaweed. This research aimed at ascertaining nitrogen
sources has role to influence life span of compost activator and
the activity of compost activator can be improve by composting
process.
The ratio of compost activator consortium is 1:1 consist of
Aspergillus niger and Rhizobium sp. Consortium is cultured into
variation nitrogen sources medium by different concentration,
2,5% and 1,25% and repeated in 3 times. Total colony is
measured by TPC method in 6 dillutions. The protein content was
measured by using Bradford method. Then the activator compost
is applied for making compost from leaves waste as the organic
material. The degradation activity of compost activator is
analyzed by C/N ratio. The data of this research is analyzed in descriptive by arrange colony growth curve, slope of activator
compost growth, protein content, temperature of compost, and
table of C/N ratio. Based on the research, the best medium for
enhancing activator compost life span is seaweed concentration
1,25% because the slope analyze was low than the other and it
was assumed that cell total was stable based on the death phase
was low. Besides, seaweed was easily to maintain for making
medium growth of compost activator and cheap. Then, the best
activator compost for C/N ratio was determined in seaweed
medium with the result was 14,15/1.
