Abstract

Penelitian berikut mengenai pengidentifikasian asam
lemak yang terdapat pada spesies Rhodotorula graminis NBRC
0190 pada National Institute of Technology and Evaluation
Jepang. Ragi R. graminis dibiakkan pada media padat dan media
cair berupa campuran glukosa, pepton dan ekstrak yeast
(1:0,5:0,3). Selanjutnya ragi diinokulasi selama rentang waktu
tertentu sesuai data kurva pertumbuhan. Biomassa hasil inokulasi
kemudian dibersihkan dan dikeringkan selama 24 jam. Sel kering
diambil lipid totalnya dengan larutan kloroform-metanol (2:1).
Lipid total selanjutnya ditransesterifikasi agar dapat diketahui
kandungan asam lemaknya dengan metode KLT dan KG-SM.
Hasil penelitian yang diperoleh menunjukkan bahwa pada plat
KLT dengan eluen petroleum eter:dietil eter:etil asetat (3:3:2)
terdapat perubahan senyawa lipid menjadi senyawa metil ester
setelah mengalami reaksi transesterifikasi serta adanya delapan
asam lemak yang muncul pada kromatogram KG. Kedelapan
asam lemak tersebut yaitu metil heksadekanoat, asam z-7-
heksadekenoat; asam n-heksadekanoat; metil oktadek-9-enoat,
metil 14-metil-heptadekanoat, asam oleat, asam oktadek-9-enoat;
dan bis(2-etilheksil) adipat yang telah umum ditemukan pada R.
graminis lainnya

The following studies regarding the identification of
fatty acids contained in the species Rhodotorula graminis NBRC
0190 Laboratory at the National Institute of Technology and
Evaluation Japan. Yeast R. graminis cultured on solid media and
liquid media in the form of a mixture of glucose, peptone and
yeast extract (1:0.5:0.3). Furthermore, the yeast inoculated during
a certain time span for which data growth curve. Biomass results
of inoculation is then cleaned and dried for 24 hours. Dry cell
taken total lipid with a solution of chloroform-methanol (2: 1).
The next total lipid transesterified in order to know the fatty acid
content by using TLC and GC-MS. The results obtained show
that the TLC plate with eluent petroleum ether:diethyl ether:ethyl
acetate (3:3:2) there are changes in lipid compound into methyl
ester compound after a transesterification reaction as well as the
presence of eight fatty acid that appears on the chromatogram
GC. These 8th fatty acids, namely hexadecanoic acid, methyl
esters; z-7-hexadecenoic acid, n-hexadecanoic acid, 9-
octadecanoic acid, methyl esters; heptadecanoic acid, 14-methyl-,
methyl esters; octadec-9-enoic acid; and hexanedioic acid, bis(2-
ethylhexyl) ester which is commonly found on the others R.
graminis.
