Abstract

Jeruk JC (Japansche Citroen) merupakan salah satu jenis jeruk yang sering digunakan sebagai stok batang bawah. Batang bawah harus mampu tumbuh dan beradaptasi pada kondisi lingkungan yang kurang baik, salah satunya terhadap cekaman salinitas. Tujuan penelitian ini yaitu untuk mengetahui pengaruh media dengan konsentrasi NaCl yang berbeda terhadap pertumbuhan kalus jeruk JC (Japansche Citroen) secara in vitro. Eksplan berupa kalus jeruk JC diinokulasikan pada media MS cair dengan penambahan NaCl konsentrasi 0 mM, 50 mM, 100 mM, dan 150 mM. Parameter pengamatan yang digunakan yaitu pengamatan warna kalus, berat basah kalus, viabilitas kalus, struktur mikroskopis kalus, dan struktur makroskopis kalus. Data dianalisis menggunakan ANOVA One Way dan dilanjutkan dengan uji Tukey pada taraf kepercayaan 95%. Berdasarkan hasil penelitian ini konsentrasi NaCl berpengaruh terhadap pertumbuhan kalus jeruk JC. Pada konsentrasi 150 mM kalus jeruk JC dapat bertahan hidup terhadap kondisi salinitas tinggi. Berat basah kalus tertinggi yaitu pada perlakuan kontrol sebesar 0,876 g, dengan tekstur remah serta kalus berwarna putih kehijauan. Berat basah terendah yaitu sebesar 0,573 g pada perlakuan NaCl konsentrasi 150 mM, dengan tekstur intermediet serta kalus berwarna putih kekuningan. ===================================================================================================== JC (Japansche Citroen) is one type rootstock of citrus. Rootstock should be able to grow and adapt to the adverse environmental conditions, one of which is against salinity stress. The purpose of this study is to determine the effect of media with different concentrations of NaCl on the growth of callus JC (Japansche Citroen) in vitro in order to obtain an JC callus will be the rootstock resistant to salinity stress. The explants used are JC callus, callus was inoculated on liquid MS medium with the addition of NaCl concentration from 0 mM, 50 mM, 100 mM and 150 mM. The observation parameter used is observation of the color of callus, callus fresh weight, viability of callus, microscopic structure callus, and macroscopic structure callus. Data were analyzed using ANOVA One Way and continued by Tukey with p value < 0,05. The results of this research the concentration of NaCl effect on the growth of orange JC callus. At concentration of 150 mM orange callus JC can survive against high salinity conditions. The highest wet weight of callus is on the control treatment of 0.876 g, with friable texture and whiteish-green callus. The lowest wet weight of 0.573 g in the treatment of NaCl concentration of 150 mM, with intermediate texture and yellowish-white callus.
