Abstract

Propolis merupakan hasil produksi dari lebah madu yang telah diketahui mempunyai banyak aktivitas biologis seperti antibakteri, antiinflamasi, antikanker dan antioksidan. Trigona sp., merupakan salah satu spesies lebah tak bersengat yang dapat menghasilkan propolis dengan kualitas baik. Tujuan dari dilakukannya penelitian ini adalah untuk mengetahui jenis pelarut yang paling efektif diantara metanol, etanol 70%, dan etil asetat dalam mengekstrak senyawa-senyawa fenolik dan flavonoid yang terdapat dalam propolis, serta untuk mengetahui nilai kandungan fenolik, flavonoid dan aktivitas antioksidan dari ekstrak propolis. Berdasarkan hasil analisa kandungan fenolik, dan flavonoid diketahui bahwa pelarut yang paling efektif dalam mengekstrak senyawa-senyawa fenolik dan flavonoid dalam propolis adalah pelarut metanol. Hasil analisa kandungan fenolik total diperoleh nilai kandungan fenolik dalam ekstrak propolis metanol (EPMe) sebesar 498,925 ± 6,322 µg Asam Galat Ekivalen (AGE)/g sarang lebah kering, ekstrak propolis etil asetat (EPEA) sebesar 140,302 ± 9,541 µg (AGE)/g sarang lebah kering, ekstrak propolis etanol 70% (EPEt) sebesar 104,628 ± 0,583 µg (AGE)/g sarang lebah kering. Hasil analisa kandungan flavonoid total diperoleh kandungan flavonoid pada EPMe sebesar 129,265 ± 2,166 Kuersetin Ekivalen (KE)/g sarang lebah kering, EPEA sebesar 12,088 ± 0,553 µg (KE)/g sarang lebah kering, EPEt sebesar 0,405 ± 0,064 µg (KE)/g sarang lebah kering. Nilai aktivitas antioksidan disajikan dalam bentuk persentase (%) dekolorasi. Nilai aktivitas antioksidan dari ketiga ekstrak propolis adalah 90,697 ± 0,673% (EPMe), 90,354 ± 0,657% (EPEA) dan 88,782 ± 0,098% (EPEt).

Propolis is honeybees product which has been known has many biological activities, such as antibactery, antiinflamation, anticancer, and antioxidant. Trigona sp., one of stingless honeybees species that produce well qualities propolis. The aim of this research are to know the best solvent between methanol, ethanol 70% and ethyl acetate to extract phenolic and flavonoid compounds and to know the value of phenolic, flavonoid contents, and antioxidant activity of propolis extracts. Based on results from total phenolic and flavonoid content assays, the best solvent to extract phenolic and flavonoid content of propolis is methanol. The results of total phenolic content assay from methanolic extract propolis (MEP) was 498,925 ± 6,322 µg Gallic Acid Equivalent (GAE) /g dry beehives, ethyl acetate extract propolis (EAEP) was 140,302 ± 9,541 µg (GAE)/g dry beehives and ethanolic extract propolis (EEP) was 104,628 ± 0,583 µg GAE) /g dry beehives. The results of total flavonoid content from MEP was 129,265 ± 2,166 Quercetin Equivalent (QE)/g dry beehives, EAEP was 12,088 ± 0,553 µg (QE)/g dry beehives and EEP was 0,405 ± 0,064 µg (QE)/g dry beehives. Antioxidant activity of propolis was represented by percent of decoloration (% decoloration). Persent of decoloration of MEP was 90,697 ± 0,673%, EAEP was 90,354 ± 0,657% and EEP was 88,782 ± 0,098%.
