Abstract

Kedelai (Glycine max L.) merupakan komoditas pangan
utama selain padi dan jagung yang mengandung protein, minyak, dan karbohidrat tinggi. Faktor dominan yang berpengaruh pada rendahnya produksi tanaman kedelai adalah perubahan iklim terutama cekaman genangan. Pada penelitian ini, induksi variasi dilakukan dengan metode iradiasi sinar Gamma dengan dosis penyinaran 25Gy, 50Gy, 75Gy, dan 100Gy. Varian hasil iradiasi kemudian diseleksi pada kondisi tergenang dengan konsentrasi air sebesar 100%, 150%, 200%, dan 250%. Parameter pertumbuhan yang diamati antara lain bintil akar, akar adventif, jumlah akar, bobot basah tanaman, dan bobot kering tanaman. Sedangkan pengamatan perubahan karakter genotip dilakukan dengan pendekatan Inter Simple Sequence Repeats (ISSR). Interaksi antara Iradiasi dan Genangan memberikan pengaruh yang signifikan pada parameter jumlah bintil akar, jumlah akar, bobot basah dan kering tanaman, namun tidak signifikan pada parameter akar adventif. Hasil pengamatan dengan marka molekuler ISSR menunjukkan polimorfisme pada perlakuan iradiasi dan primer yang informatif adalah primer ISSR-1, ISSR- 3, ISSR-5.

Soybean (Glycine max L.) is the main food commodity
besides rice and maize which contain high protein, oil, and
carbohydrate. The dominant factor that influences the low
production of soybean crop is the climate change. In this study, induction of variation was done by irradiation method of Gamma rays with radiation dose 25Gy, 50Gy, 75Gy, and 100Gy. The irradiation variant was then selected under waterlogging conditions with 100%, 150%, 200%, and 250% water concentrations. The observed growth parameters were root nodule, adventif root, root number, wet weight of plant, and dry weight of plant. The observation of genotype character is done by Inter Simple Sequence Repeats (ISSR) technique. The interaction between irradiation and inundation had a significant influence on the parameters of root nodules, number of roots, wet and dry weight of plants, but not significantly on adventitious root
parameters. Observations with molecular markers of ISSR
showed polymorphisms in irradiation treatments and informative primers were ISSR-1 primers, ISSR-3, ISSR-5.
