Abstract

Nyamplung (Calophyllum inophyllum) merupakan salah
satu jenis tanaman non-pangan yang mempunyai nilai ekonomi
yang tinggi, karena hampir seluruh bagian tanamannya (batang,
daun, bunga, biji, dan getah) dapat menghasilkan bermacam
produk yang dapat dimanfaatkan untuk berbagai kebutuhan hidup
manusia. Pada berbagai bagian dari tanaman nyamplung
mengandung beberapa komponen bioaktif misalnya kumarin dan
xanthone. Dari penelitian yang dilakukan Laure et al., 2008,
senyawa kumarin menunjukkan aktivitas anti HIV yang termasuk
ke dalam NNRTI (Non-Nucleoside Reverse Transcriptase
Inhibitor). Penelitian ini bertujuan untuk mengambil ekstrak
kumarin dan xanthone, mengetahui kadarnya dari minyak
nyamplung, dan mengetahui metode untuk pemurniannya.
Minyak nyamplung diekstraksi sebanyak 8 stage, rasio pelarut
terhadap minyak 5:1 (g/g), dan rasio pelarut PE-methanol 98%
75:25 (g/g). Fase methanol dikumpulkan dan diuapkan untuk
mendapatkan fraksi lemak polar (Polar Lipid Fraction, PLF).
Selanjutnya menerapkan metode stirred batched-wise dengan
rasio PLF/silica gel = 1:2; 1:3; dan 1:6 (g/g), rasio n-hexane/PLF
= 25 g/g, pada suhu 20°C. Campuran tersebut diaduk pada
kecepatan 1000 rpm selama 10 menit sampai beberapa batch dan
terpisahnya kumarin dan xanthone. Fraksi xanthone dan kumarin
diuji dengan TLC dan spektrofotometer UV-VIS. Kadar kumarin
terbesar ada pada fraksi metanol perbandingan 1:6 sebesar 2,56%; sedangkan kadar xanthone terbesar di fraksi n-hexane
perbandingan 1:6 sebesar 3,94%.

Nyamplung (Calophyllum inophyllum) is one of nonediable
plant that has high economic value, since almost every
parts of the plant (stems, leaves, flowers, seeds, and gums) can
produce a variety products that can be used for various human life
needs. In various parts of nyamplung contains several bioactive
compounds, for example coumarin and xanthone. From research
conducted by Laure et.al., 2008, coumarin showed anti-HIV
activity that belongs to the NNRTI (Non-Nucleoside Reverse
Transcriptase Inhibitor). This research aims to extract coumarin
and xanthone, determine the levels of those components in the
crude oil so, and determine the methods to purify them.
Nyamplung oil extracted 8 times, the solvent to oil ratio is 5:1
(g/g), and PE-methanol 98% solvent ratio is 75:25 (g/g).
Methanol phase was collected and evaporated to obtain polar lipid
fraction (PLF). Furthermore, the stirred batched-wise method was
applied with the ratio of PLF/silica gel = 1:2; 1:3; and 1:6 (g/g)
and the ratio of hexane/PLF = 20 g/g at the temperature 20°C.
The mixture is then stirred at 1000 rpm for 10 minutes until a few
batches and coumarin and xanthone obtained. Coumarin and
xanthone fraction then being analysed using TLC and UV-VIS
spectrophotometer. The biggest content of coumarin found in
methanol fraction of ratio 1:5 around 2,56%; in the other hand the
biggest content of xanthone found in n-hexane fraction of ratio
1:6 around 3,94%.
