Abstract

Lipase (EC 3.1.1.3) adalah enzim kelas hidrolase, yang
bekerja dalam hidrolisis triasilgliserol menjadi asam lemak dan
gliserol. Lipase dapat diproduksi dari mikroorganisme. Salah
satu kelompok mikroorganisme yang mampu memproduksi lipase
adalah marine yeast.
Produksi lipase dari marine yeast dapat dilihat dengan uji
aktivitas lipase. Produksi lipase dari marine yeast dipengaruhi
oleh beberapa faktor kondisi lingkungan, antara lain masa
inkubasi, suhu, dan pH. Penelitian ini bertujuan mencari variasi
kondisi lingkungan optimal untuk produksi lipase dari Candida W
3.8
Penelitian ini menggunakan Candida W 3.8 yang telah
diremajakan, kemudian diinkubasi selama 48 jam pada suhu
ruang. Isolat diinokulasi ke dalam media Yeast Malt Broth
(YMB) untuk pembuatan kurva pertumbuhan. Kultur dimasukkan
ke dalam media produksi dengan variasi kondisi lingkungan yang
terdiri dari masa inkubasi serta pH dan suhu. Selanjutnya diuji
aktivitas lipase dengan menggunakan p-Nitrophenyl Palmitate
(pNPP). Absorbansi diukur dengan spektrofotometer pada
panjang gelombang 410 nm. Hasil absorbansi dimasukkan ke
dalam persamaan kurva standar p-Nitrophenol (pNP),
selanjutnya dimasukkan ke dalam rumus aktivitas lipase. Nilai
yang diperoleh dinyatakan dalam satuan aktivitas enzim yaitu
U/ml. Desain penelitian menggunakan kombinasi dua faktor.
Data yang diperoleh dianalisis secara deskriptif kuantitatif

Hasil penelitian didapatkan kondisi lingkungan optimal
untuk produksi lipase oleh Candida W 3.8 adalah pada masa
inkubasi 120 jam, serta rentang suhu 27⁰C - 45⁰C dan pH 7

Lipase (EC 3.1.1.3) is a hidrolase class enzyme, which works
in triglyceride hydrolisis into fatty acid and glycerol. Lipase can
be produced from microorganism, one of which is marine yeast.
Lipase production from marine yeast can be seen in lipase
activity test. It is affected by several environmental factors, such
as pH, temperature, and incubation time. This research intends to
look for the optimization variation on those environmental factors
combination to produce lipase from Candida W 3.8
This research is conducted using rejuvenated Candida W 3.8
which was incubated for 48 hours in room temperature. Then the
isolate were inoculated in media Yeast Malt Broth (YMB) in
order to create growth curve. The culture then were being put
into media production with various change in incubation time,
pH, and temperature. Lipase activity were tested by using p-
Nitrophenyl Palmitate (pNPP). Absorbency was measured by
spektrofotometer on wavelength 410 nm. Absorbency result were
put into equation of standard curve p-Nitrophenol (pNP), and
then being used as an input on lipase activity formula. The result
are stated in enzym activity unit, which is U/ml. Research design
is being done by the combination of two factors, and the obtained
data analyzed by using descriptive quantitative method.
The optimum environment in order to produce lipase enzyme
by using Candida W 3.8 is on 120 hours incubation time,
temperature range in 27⁰C - 45⁰C and pH 7
