Abstract

Formulasi pembawa obat untuk sistem penghantaran obat
Mengapung (Floating Drug Delivery Systems) berbasis Kalsium
Karbonat, HPMC K15M dan Sodium Alginat telah dilaksanakan.
Tujuan penelitian ini adalah untuk mengetahui pengaruh rasio
material Drug Carrier (CaCO3, HPMC K15M, dan Sodium
Alginat) terhadap floating behavior dan release kinetics. Serbuk
kalsium karbonat dihasilkan dari ekstrak cangkang kerang hijau
dan Kulit Telur. Sodium alginate digunakan sebagai crosslinking
dengan ion Kalsium dan HPMC K15M digunakan sebagai
peningkat bioavailabilitas obat. Obat yang digunakan adalah
Aluminium Hidroksida sebagai obat lambung. Setiap formulasi
dilakukan pengujian floating behavior untuk mengetahui waktu
lag bouyancy dan bouyancy duration serta pengujian In vitro
Dissolution untuk mengetahui konsentrasi pelepasan obat dan
mekanisme pelepasan. Bermacam variabel formulasi seperti
konsentrasi Kalsium Karbonat, HPMC K15M dan Sodium
Alginat secara signifikan berpengaruh terhadap floating behavior
dan pelepasan obat in vitro. Analisa untuk pola pelepasan obat
pada tablet seperti Marshmallow menunjukan bahwa pelepasan
obat mengikuti persamaan model First-order.

Formulations of the drug carrier for Floating Drug
Delivery Systems based on Calcium Carbonate, HPMC K15M,
and Sodium Alginate were implemented. The purpose of this
study was to determine the influence of Drug Carrier materials
(CaCO3, HPMC K15M, and Sodium Alginate) to floating
behavior and release kinetics. Calcium Carbonate powder
produced from the extract of green mussel shells and eggshells.
Sodium Alginate was used as a crosslinking with calcium ions
and HPMC K15M was used as a drug bioavailability enhancer.
The drug used was Aluminium Hydroxide as stomach medicine.
Each formulation was tested to determine floating behavior such
as lag bouyancy time and bouyancy duration and In Vitro
Dissolution Testing to determine the concentration of drug release
and release mechanism. Various formulation variables such as the
concentration of Calcium Carbonate, Sodium Alginate, and
HPMC K15M significantly affected the floating behavior and In
Vitro drug release. Analysis of the release pattern on the
Marshmallow-like tablet showed that the drug release followed
the First-order model.
