Abstract

Optimasi amobilisasi bromelin dengan matriks pendukung kitosan telah diteliti. Enzim bromelin yang diisolasi dari buah nanas mentah maupun matang diperoleh aktivitas tertinggi pada pengendapan 40% dengan ammonium sulfat jenuh. Kondisi optimum amobilisasi enzim bromelin didapatkan pada konsentrasi kitosan 240,4 mg dan bromelin 10 mg dengan jumlah enzim teramobil 9,5417 mg dan aktivitas dalam 10 mg enzim amobil yang diuji adalah 86,2067 Unit. Uji aktivitas enzim amobil terhadap pengaruh substrat kasein didapatkan aktivitas tertinggi pada kasein 3000 ppm. Uji perulangan enzim amobil dapat digunakan hingga enam kali dengan efisiensi 57,69%
=======================================================================================Optimization of immobilization of bromelain using support matrix chitosan had been investigated. Bromelain enzyme which had been isolated from raw pineapple and ripe pineapple obtained the highest activity at 40% precipitation with ammonium sulfate saturation. Optimum condition of immobilization bromelain was on 240,4 mg chitosan concentration and 10 mg bromelain concentration with restrained enzyme and activity was 9,5417 mg and 86,2067 Unit. Activity assay of bromelain immobilized to the effect of casein substrate obtained the highest activity on 3000 ppm casein concentration. Repetition assay of bromelain immobilized can be used until six times reusable with 57,69% efficiency
