Abstract

Optimalisasi biodegradasi DDT oleh Pleurotus eryngii dengan
penambahan bakteri telah dilakukan, dimana bakteri yang digunakan adalah
Pseudomonas aeruginosa, Bacillus subtilis dan Ralstonia picketti. Screening
dilakukan dengan menambahkan masing-masing bakteri (7 ml) ke dalam kultur P.
eryngii dan konfrontasi assay jamur dan bakteri. R. picketti menunjukkan
sinergitas dengan P. eryngii dan digunakan dalam proses degradasi selanjutnya. R.
pickettii ditambahkan sebesar 1, 3, 5, 7 dan 10 ml (1 ml ≈ 1,44 x 1013 R. pickettii
sel/ml kultur) kedalam 20 ml kultur P. eryngii. Degradasi DDT tertinggi
ditunjukkan pada penambahan bakteri 7 ml dengan jumlah degradasi sebesar
78,34 %. Degradasi DDT pada waktu penambahan bakteri hari ke-0, 1, 3 dan 5
secara berurutan adalah sebesar 78%, 70%, 80%, dan 38%. Produk metabolit yang
dihasilkan selama proses degradasi adalah DDD, DDE dan DDMU

Optimization of biodegradation of DDT by Pleurotus eryngii with the
addition of bacteria, had been investigated. Pseudomonas aeruginosa, Bacillus
subtilis and Ralstonia picketti were screened by adding each of them (7 mL) in P.
eryngii culture and also based on fungi and bacteria confrontation assay. R.
picketti showed synergism with P. eryngii, which used for further experiment by
adding to P. eryngii culture at 1, 3, 5, 7 and 10 ml (1 ml ≈ 1.44 x 1013 R pickettii
cell/ mL culture). The addition of 7 mL of R. pickettii showed the highest DDT
degradation about 78.34%. The addition of 7 ml of R. pickettii on days 0, 1, 3, and
5 showed DDT degradation approximately 78%, 70%, 80% and 38%,
respectively. DDD, DDE and DDMU were detected as metabolite products from
DDT degradation by P. eryngii and R. pickettii.
