Abstract

Telah dilakukan penelitian screening tujuh minyak nabati untuk minyak
Imersi Mikroskop Optik. Screening dilakukan dengan cara, minyak nabati diekstrak
dengan teknik hydrolik press dan sokletasi, minyak kasar kemudian dimurnikan
dengan metode degumming, netralisasi, bleaching, sentrifugasi, winterisasi,
dikarakterisasi dengan Uv-Vis dan FTIR, pengukuran parameter minyak imersi,
dan dilakukan pengujian pada Mikroskop Optik. Data yang diperoleh kemudian
dibandingkannya dengan data hasil pengukuran dan pengujian minyak imersi
standar. Hasil ekstrak dan pemurnian menghasilkan perubahan warna dan
menambah kejernihan minyak, namun masih mengandung asam lemak bebas yang
cukup tinggi, yaitu berkisar dari 0,460 sampai 0,740 mg KOH/g dibanding minyak
imeri standar hasil uji yaitu 0,126 mg KOH/g. Hasil karaterisasi dengan FTIR dan
Uv-Vis, teridentifikasi gugus fungsi minyak nabati mirip dengan minyak standar,
yaitu kedua kelompok minyak ini memiliki gugus –OH, C – H, C = O, C – O, dan
juga terdapat kesamaan bilangan dispersi dengan minyak standar yaitu pada kisaran
panjang gelombang pendek. Hasil analisis parameter fisika dan kimia memberikan
kesimpulan bahwa setiap parameter memberikan pengaruh yang variatif terhadap
besarnya nilai apertur (NA) dan fotograph hasil pengamatan. Parameter yang
berpengaruh nyata adalah viskositas, indeks bias, dan bilangan asam. Hasil
perhitungan Nilai Apertur (NA) minyak kenari (Canarium commune / C. Avenue)
> minyak ketapang (Terminalia Catapa) = minyak kesambi (Scheichera oleosa) =
minyak kacang tanah (Arachis Hypogaea L) > minyak nyamplung (Calophyllum
Inophyllum) > minyak Kepuh (Sterculia Foetida L) > minyak Jarak kepyar (Ricinus
Communis. L). Pengujian pada mikroskop optik tipe Stereo Cahaya Binokuler,
merk leybold Didactic GmbH, dengan hasil pengamatan lebih jelas adalah minyak
kenari, disusul kacang tanah, ketapang, kesambi, kepuh, nyamplung, dan jarak
kepyar. Hasil pengujian disimpulkan bahwa lima dari tujuh minyak nabati bisa
dijadikan alternatif minyak imersi mikroskop optik

It has been conducted a research about screening of seven variants of vegetable oil
for immersion oil in Optical Microscope. Screening is done by extracted the
vegetable oil using hydraulic press and soxhletation technique , the crude oil is then
purified by degumming, neutralization, bleaching, centrifugation, winterization
method and characterized with Uv-Vis and FTIR for measuring the parameters of
immersion oil, and it tested on Optical Microscope. The result then compared with
the data standard immersion oil. The result from extraction and purification produce
a color change and the purity of the oil is increased, but it still contains free fatty
acids and it’s quite high with the range from 0.460 to 0.740 mg KOH / g, compared
to the standard Immersion oil test results is 0.126 mg KOH / g. The results
characterized with FTIR and Uv-Vis, it is identified that the functional groups of
vegetable oils are similar to the standard oil, which is both oil has -OH groups, C -
H, C = O, C - O, and also there are similarities with standard oil dispersion numbers,
they are both located in the range of short wavelengths. From the results of the
analysis of physical and chemical parameters, it can be concluded that each
parameter varied effect on the value of the aperture (NA) and phootograph
observations. The significant parameters are the viscosity, refractive index, and
acid. The results of calculation of value Aperture (NA) walnut oil (Canarium
commune / C Avenue) > oil Ketapang (Terminalia Catapa) = oil kesambi
(Scheichera oleosa) = oil peanut (Arachis hypogaea L) > nyamplung oil
(Calophyllum inophyllum) > oil Kepuh (Sterculia foetida L) > Distance kepyar oil
(Ricinus communis, L). The testing using an optical microscope Binocular Light
Stereo type, brand Leybold Didactic GmbH, gives result that the obvious
observation isobtained by using, followed by peanuts, ketapan, kesambi, billowing,
nyamplung, and castor. It can be concluded that five of the seven vegetable oils
can be used as alternatives for immersion oil on optical microscopy
