Abstract

Seiring perkembangan ilmu pengetahuan dan teknologi, plastik merupakan salah satu polimer sintetik yang mengalami peningkatan dalam penggunaannya. Penggunaan plastik berupa kantong kresek hasil daur ulang dengan berbagai warna sangat diminati oleh masyarakat. Sifat plastik yang tidak mudah terdegradasi di alam mengakibatkan masalah lingkungan.
Penelitian ini dilakukan untuk mengisolasi dan mengkarakterisasi bakteri tanah sampah yang mampu mendegradasi plastik secara biokimia. Uji Biodegradasi plastik ditinjau dari prosentase kehilangan berat kering, pengukuran densitas sel biofilm, densitas sel kolom air dan pH tiap bulan selama 4 bulan masa inkubasi.
Hasil penelitian didapatkan inokulum mikroorganisme yang ada di tanah sampah mampu mendegradasi plastik yang ditunjukkan dengan prosentase kehilangan berat kering plastik putih bening rata-rata per bulan sebesar 1% dan hitam sebesar 1,87%. Berdasarkan kunci dikotomi Bergey’s Determinative of Bacteriology isolat tersebut cenderung masuk ke dalam Genus Bacillus (PPs 9 dan PPs 11), Corynebacterium (PPs 7), Lactobacillus (PPs 2), Vibrio (PPs 5, PPs 8 dan PPs 10), Aeromonas (PPs 1, PPs 4, PPs 6, dan PPs 12), Yersinia (PPs 13) dan Neisseria (PPs 3)

As the development of science and technology, plastic is one of the synthetic polymer that have increased in use among the community. The use of plastics in the form of recycled plastic bags with various color are in great demand by the public. Plastic properties that are not easily degraded in nature resulting in environmental problems.
This research was conducted to isolate and characterize waste soil bacteria capable of degrading plastic. Biodegradation test of plastic in terms of percentage loss of dry weight, measurement of biofilm cell density, measurement of water column cell density and pH every month for 4 month of incubation.
The results showed that the inoculum in the soil microorganisms capable of degrading plastic bins with a dry weight percentage loss of translucent white plastic per month on average by 1% and amounted to 1.87% black. Based on the key dichotomy Bergey's Determinative of Bacteriology isolates tend to enter into the genus Bacillus (PPS 9 and PPS 11), Corynebacterium (PPS 7), Lactobacillus (PPs 2), Vibrio (PPs 5, PPs 8 and PPs 10), Aeromonas (PPs 1, 4 PPs, PPs 6, and PPs 12), Yersinia (PPS 13) and Neisseria (PPS 3).
