Abstract

Penelitian ini bertujuan untuk meningkatkan bioavailabilitas dan kelarutan kandungan bioaktif pada ekstrak C. xanthorrhiza sehingga dapat diterapkan pada
industri farmasi. Curcuma xanthorrhiza Roxb (C. xanthorrhiza) dikenal sebagai Temulawak adalah spesies tanaman yang termasuk dalam kelompok Zingiberaceae dan merupakan tanaman andalan asli Indonesia yang memiliki banyak manfaat dalam bidang kesehatan. Pada pengobatan tradisional, C. xanthorrhiza dapat digunakan sebagai penanganan hepatitis, diabetes, hipertensi, hilangnya nafsu makan, dan beberapa penyakit lainnya. Hal ini didukung oleh adanya isolasi kandungan yang ada di dalamnya. Ekstrak dari C. xanthorrhiza mengandung diarylheptanoid (kurkuminoid) dan terpenoid (xanthorrhizol) terutama sesquiterpenes. Namun C. xanthorrhiza memiliki bioavailabilitas dan kelarutan yang rendah di dalam air sehingga menghambat pemanfaatannya. Sehingga dalam peneilitian ini, ekstrak dari C. xanthorrhiza dimikronisasi dengan menggunakan metode superkritis sebagai anti-solvent (SAS). Adanya penambahan biopolimer PVP diharapkan dapat meningkatkan kelarutan dari ekstrak C. xanthorrhiza dalam aqueous. Ekstrak C. xanthorrhiza diperoleh dengan menggunakan metode soxhletasi kemudian dilanjutkan ke proses mikronisasi partikel dengan menggunakan metode SAS. Fluida superkritis yang digunakan sebagai anti-solvent pada penelitian ini adalah Karbondioksida (CO2). Jenis Pelarut yang digunakan untuk melarutkan ekstrak C. xanthorrhiza pada penelitian ini adalah Aseton, Etil Asetat, dan Diklorometan. Konsentrasi solution adalah 2 mg/mL. Flowrate solution dan anti-solvent sebesar 0.25 mL/menit dan 15 mL/menit. Suhu operasi konstan pada 40°C. Tekanan operasi divariasikan pada 8 Mpa, 12 MPa, dan 16 MPa. Sedangkan ratio ekstrak C. xanthorrhiza/PVP bervariasi yaitu 1:0, 1:10, dan 1:20. Mikropartikel yang terbentuk kemudian dianalisa menggunakan SEM, FTIR, dan Spektrofotometer UV-VIS. Hasil optimum dengan ukuran partikel sebesar ±180.11 nm diperoleh pada variabel tekanan operasi 12 MPa, ratio C. xanthorrhiza/PVP 1:10, dan jenis pelarut Etil Asetat. Simulasi menggunakan Computational Fluid Dynamic (CFD) bertujuan untuk mengetahui profil aliran fluida di dalam nozzle. Dengan adanya simulasi CFD mengenai aliran internal di dalam nozzle diharapkan dapat berguna dalam menentukan parameter operasi yang lebih optimum.

This research was aimed to increase the bioavailability and solubility of bioactive compound in C. xanthorrhiza extract so it can be applied to the pharmaceutical industry. Curcuma xanthorrhiza Roxb (C. xanthorrhiza) known as Temulawak is a plant species belonging to the group Zingiberaceae and is an original plant from Indonesia that has many benefits for people’s health. In traditional medicine, C. xanthorrhiza can be used as a treatment for hepatitis, diabetes, hypertension, loss of appetites, and some other diseases. Extract from C. xanthorrhiza contain diarylheptanoids (curcuminoids) and terpenoids (xanthorrizol) especially sesquiterpenes. However, C. xanthorrhiza has low bioavailability and solubility in the water, thus inhibiting its utilization. Therefore, in this research, extract from C. xanthorrhiza was micronized using Supercritical Fluid as Anti-Solvent (SAS) method. The addition of biopolymer PVP was expected to increase the solubility of C. xanthorrhiza extract in aqueous solution. The extract of C. xanthorrhiza was obtained by using soxhletation then continued to the process of particle micronization using SAS method. The supercritical fluid used as anti-solvent in this study was Carbon dioxide (CO2). Acetone, Ethyl Acetate, and Dichloromethane were used as solvent to dilute C. xanthorrhiza extract. Solution concentration was 2 mg/mL. The flowrate of solution and anti-solvent were constant at 0.25 mL/min and 15 mL/min. Operation temperature was kept at 40°C. Operation pressure was varied from 8 MPa to 16 MPa. Meanwhile the extract C. xanthorrhiza/PVP ratio varied at 1:0, 1:10, and 1:20. The formed microparticles were analyzed using SEM, FTIR, and Spektrofotometer UV-VIS. The optimum result with particle size ±180.11 nm was obtained at operating pressure 12 MPa, C. xanthorrhiza/PVP ratio at 1:10, and Ethyl Acetate as the solution solvent. Simulation using Computational Fluid Dynamic (CFD) aimed to determine the fluid flow inside the nozzle. With CFD simulation of internal flow inside nozzle was expected to determine the optimum operating parameters.
