Abstract

Early screening of Inherited Metabolic Diseases is important to avoid DNA damage caused by them. Maple Syrup Urine Disease, an Inherited Metabolic Disease, has in leucine a specific marker found in body fluids, such as urine. Specific wavelengths are associated to specific markers. Spectrometry has proven to be useful in identifying specific metabolites and markers. The purpose of this study was to develop an initial approach to an affordable portable point-of-care diagnostic prototype device for early screening of Maple Syrup Urine Disease using spectrometry. To accomplish this, the component selection, framework design, and redesign, to improve the performance of the system, and a neural network approach, using the data captured by the device, were studied. The designed prototype can differentiate similar liquids, including artificial urine with different concentrations of leucine, being ideal to start a research line to improve early non-invasive diagnostic devices. With a principal component analysis approach, the device was able to identify different liquids with a 97.61% of accuracy, using the first two components. The developed neural network for the device, was able to identify artificial urine with low concentrations of leucine with an accuracy of 83.33% with test data acquired with the device. Experimental tests showed an accuracy of 76.67%, with a sensitivity of 83.33%, and a specificity of 72.22%. Making the developed system a reliable tool to identify artificial urine samples with normal and elevated leucine level values.
