Abstract

Yeast laut memiliki keunggulan bila dibandingkan dengan terrestrial yeast, diantaranya adalah memiliki toleransi osmotik yang tinggi dan produktivitas enzim dengan karakter yang unik. Enzim mengambil peran dalam proses degradasi, termasuk oksidasi terminal, pemecahan rantai dan metabolisme asam lemak yang menyebabkan menurunnya berat molekul dan hidrofobisitas polimer. Plastik High Density Polyethylene mengandung karbon dan hidrogen dengan sedikit percabangan yang memberikan gaya antarmolekul lebih kuat dan kekuatan tarik (±4550 psi) lebih besar dari pada Low Density Polyethylene. Produksi enzim yeast dilakukan selama 7 hari pada Mineral Salt Medium Broth (MSMB) dengan potongan plastik High Density Polyethylene (HDPE). Parameter uji enzimatik dilakukan secara kualitatif dan kuantitatif. Hasil penelitian uji kualitatif menunjukkan 2 jenis isolat menghasilkan enzim mangan peroksidase dan alkana hidroksilase, sedangkan 1 jenis isolat lainnya menghasilkan enzim lipase dan alkana hidroksilase.

Marine yeast has advantages compared to terrestrial yeast, including its high osmotic tolerance and enzyme productivity with unique characteristics. Enzymes take a role in degradation process, including terminal oxidation, chain breakdown and fatty acid metabolism leading to decreased molecular weight and polymer hydrophobicity. High Density Polyethylene contains carbon and hydrogen with less branching which provides stronger intermolecular forces and tensile strength (± 4550 psi) greater than Low Density Polyethylene. Enzyme production was carried out for 7 days on Mineral Salt Medium Broth (MSMB) with pieces of High Density Polyethylene (HDPE) plastic. The enzymatic test parameters were carried out qualitatively and quantitatively. The results of the qualitative test showed that 2 isolates produced manganese peroxidase and alkane hydroxylase enzymes, while 1 other isolate produced lipase and alkane hydroxylase enzymes.
