Abstract

Database khamir laut pendegradasi plastik dilakukan hingga tingkat spesies untuk mengetahui identitas khamir laut dan informasi terkait khamir laut pendegradasi plastik tersebut. Identifikasi isolat khamir laut pendegradasi plastik High Density Polyethylene (HDPE) dengan menggunakan marka molekuler ITS bertujuan untuk mengidentifikasi khamir laut yang berpotensi sebagai pendegradasi plastik dengan hasil yang lebih cepat dan lebih akurat. Identifikasi molekuler memiliki kelebihan yaitu lebih cepat karena tidak dipengaruhi oleh faktor internal seperti tahap pertumbuhan maupun faktor eksternal seperti lingkungan tempat tumbuh atau hidup. Metode yang digunakan dalam penelitian ini yaitu serangkaian metode molekuler seperti ekstraksi DNA genom menggunakan gDNA Yeast Kit, amplifikasi menggunakan kit MyTaqTM HS Red Mix, elektroforesis menggunakan gel agarosa, dan sekuensing DNA. Hasil penelitian menunjukkan isolat berhasil diidentifikasi dan memiliki tingkat homologi yang tinggi dengan pencarian homologi sekuen dalam database. Kedua isolat merupakan anggota dari genus Rhodotorula dan Candida. ===================================================================================================== The plastic-degrading marine yeast database was carried out at the species level to determine the identity of marine yeasts and information related to these plastic-degrading marine yeasts. Identification of high-density polyethylene (HDPE) plastic-degrading marine yeast isolates using ITS molecular markers aims to identify marine yeasts that have the potential to degrade plastics with faster and more accurate results. Molecular identification has the advantage that it is faster because it is not influenced by internal factors such as growth stage or external factors such as the environment in which it grows or lives. The methods used in this study are a series of molecular methods such as genomic DNA extraction using the gDNA Yeast Kit, amplification using the MyTaqTM HS Red Mix kit, electrophoresis using agarose gel, and DNA sequencing. The results showed that the isolates were identified and had a high level of homology by searching for sequence homology in the database. Both isolates were members of the genera Rhodotorula and Candida.
