Abstract

Tanaman Andrographis paniculata L. Ness atau sambiloto adalah jenis tanaman obat penting dari famili Acanthaceae yang banyak tumbuh di pulau Jawa Indonesia. Dalam penelitian ini potensi baru ekstrak tanaman A. paniculata sebagai agen pelindung radiasi ultraviolet (UV) telah dipelajari. Tujuan penelitian ini adalah menentukan tingkat kemampuan ekstrak A. paniculata dalam menyerap radiasi UV, mempelajari analisis fotoprotektifnya, dan mengaplikasikannya pada sediaan krim tabir surya dengan memodifikasi menggunakan nanopartikel emas hasil sintesis. Aplikasi ekstrak tanaman A. paniculata termodifikasi nanopartikel emas dilakukan pada formula tabir surya dengan komposisi tertentu. Sintesis nanopartikel emas dilakukan menggunakan metode elektrokimia dan hasilnya dikarakterisasi secara spektrofotometri ultraviolet, SEM dan TEM. Potensi baru dari ekstrak tanaman A. paniculata dipelajari pada berbagai variasi konsentrasi ekstrak dan ditunjukkan oleh nilai faktor perlindungan matahari atau Sun Protection Factor (SPF). Uji fitokimia, analisis inframerah, karakterisasi ultraviolet, identifikasi fluoresensi, analisis kromatografi cair kinerja tinggi - spektrometri massa (LCMS/MS), analisis kandungan flavonoid secara kromatografi lapis tipis (KLT) dan analisis fotoprotektif pada ekstrak tanaman A. paniculata telah dilakukan. Dari hasil penelitian diperoleh bahwa uji fitokimia menunjukkan ekstrak tanaman A. paniculata mengandung senyawa flavonoid, alkaloid, tannin, triterpenoid dan polifenol. Ekstrak tanaman A. paniculata mempunyai 2 puncak serapan maksimum ultraviolet yaitu pada panjang gelombang 230 nm dan 362 nm. Identifikasi spektrofotometri fluoresensi diperoleh puncak eksitasi ekstrak terjadi pada 300 nm (552, 11a.u) dan puncak emisi pada 605 nm (516, 02a.u). Karakterisasi secara LCMS/MS diperoleh puncak kromatogram yang mirip dengan senyawa standar quercetin-3- glikosida pada waktu retensi 2,77. Kandungan flavonoid ekstrak diperoleh sebesar 0,022 µg/mL quercetin dengan nilai Rf sebesar 0,61. Aktivitas fotoprotektif ekstrak tanaman A. paniculata memiliki nilai SPF ≥ 15. Analisis fotoprotektif ekstrak tanaman A. paniculata menghasilkan formulasi nilai SPF yang ditunjukkan dengan persamaan korelasi polinomial 1/SPF=k_1 (1/C)^2+ k_2/C+ k_3, dimana k1 = 0,0215; k2 = -0,0044 dan k3 = 0,0292. Aktivitas fotoprotektif ekstrak tanaman A. paniculata termodifikasi nanopartikel emas pada sediaan tabir surya memiliki nilai SPF sebesar 15,67 – 32,62. ====================================================================================================== The species of Andrographis paniculata L. Ness, family of Acanthaceae grows in many parts in the island of Java in Indonesia. In this research, new potentials of ultraviolet radiation protective agents from A. paniculata plant extracts were investigated. The purpose of this study was to determine the ability of A. paniculata extract modified with gold nanoparticles to absorb ultraviolet radiation in sunscreen preparations. The synthesis of gold nanoparticles was carried out using electrochemical methods and the results were characterized using ultraviolet spectrophotometry, SEM and TEM. The potentials of A. paniculata plant extracts were studied at various concentrations and shown by the value of the sun protection factor (SPF). The application of the modified gold nanoparticle extract was carried out on a sunscreen formula with a certain composition. The phytochemical test, infrared analysis, ultraviolet characterization, fluorescence identification, high performance liquid chromatography-mass spectrometry (LCMS/MS), analysis of flavonoid content by thin layer chromatography (TLC) and photoprotective formulation were performed. Based on phytochemical test, A. paniculata plant extracts contained flavonoids, alkaloids, tannins, triterpenoids and polyphenols. A. paniculata plant extracts have 2 maximum ultraviolet absorption with maximum wavelengths at 230 nm and 362 nm. The fluorescence spectrophotometry identification obtained with excitation peak at 300nm (552.11 a.u) and the emission peak at 605nm (516.02 a.u). The flavonoid content of the extracts was 0.022 µg/mL quercetin with Rf value of 0.61. The chromatogram of the LCMS/MS is similar to quercetin-3-glycoside standard at a retention time of 2.77. The photoprotective activity of A. paniculata plant extracts had SPF ≥ 15. Photoprotective analysis of A. paniculata plant extracts resulted in the formulation of the SPF value indicated by the polynomial correlation equation as follows
