Abstract

Lahan gambut dikenal sebagai lahan yang mempunyai produktivitas rendah karena kandungan unsur hara makro dan mikro sangat sedikit sehingga kurang layak digunakan sebagai lahan budidaya tanaman. Salah satu cara untuk memperbaiki kesuburan lahan gambut yaitu dengan menambahkan bakteri lignoselulolitik yang berpotensi sebagai biofertilizer. Tujuan dari penelitian ini adalah mendapatkan agen biofertilizer dari isolat indigenous lahan gambut berbahan baku Bacillus U4 dan Pseudomonas U3. Isolat yang digunakan untuk penelitian adalah Bacillus U4 dan Pseudomonas U3. Skrining penyedia NPK, IAA, uji produksi enzim amilase, selulase, dan ligninase dilakukan dengan medium selektif. Isolat dicampur dengan media pembawa serbuk gergaji, sekam, dan gambut 1 bulan. Diamati viabilitasnya dengan metode TPC. Hasil penelitian menunjukkan Bacillus U4 dan Pseudomonas U3 dapat melarutkan fosfat, memproduksi enzim amilase, selulase, lignin peroksidase, dan mangan peroksidase sehingga belum layak digunakan sebagai biofertilizer. Media pembawa yang diinkubasi 2 jam memiliki 0,96 x 105 CFU/gr dan setelah diinkubasi selama 4 minggu media pembawa terkontaminasi jamur dan CFU tidak dapat diamati dengan jelas

Peatland has low productivity since it contains negligible amounts of macro and micro nutrients for plants growth, consequently it is not suitable for plant cultivation. One of the alternative technique to increase peatlands fertility was the introduction of lignocellulolytic bacteria. The objective of this study was to obtain biofertilizer utilizing peatlands indigenous isolates which are Bacillus U4 and Pseudomonas U3. The NPK solubilizing test and IAA, amylase, cellulase, and ligninase screening test were carried out on selective media. The isolates were mixed with various carrier media such as sawdust, husk, and peat then incubated for 1 month. Viability was observed by the TPC method. The results showed that Bacillus U4 and Pseudomonas U3 were only capable to solubilize phosphate, produce amylase, cellulase, lignin peroxidase, and mangan peroxidase thus they were not suitable for biofertilizers agents. The carrier media incubated at 2 hours had 0.96 x 105 CFU/gr of viable cell and after 4 weeks of incubation the carrier medium was contaminated with fungi and CFU could not be clearly observed.
