Abstract

Skeletonema costatum merupakan mikroalga diatom yang berpotensi sebagai bahan baku biodiesel karena mengandung lipid TAG berkadar tinggi. Lipid TAG dapat diinduksi melalui perubahan jalur fotosintesis menjadi biosintesis asam lemak ketika mengalami cekaman nutrien Nitrogen dan Silika. Regulasi biosintesis asam lemak diawali oleh gen ACC1 dan ACC2. Penelitian ini bertujuan untuk mengetahui ekspresi gen ACC1 dan ACC2 dibawah kondisi cekaman Nitrogen dan Silika pada mikroalga Skeletonema costatum. Metode yang digunakan adalah analisis ekspresi gen pada sampel ekstrak RNA S. costatum dari penelitian sebelumnya menggunakan qRT-PCR.
Hasil analisis ekspresi gen menunjukkan bahwa terdapat 1 dari 4 sampel yang dapat diidentifikasi oleh qRT-PCR, yaitu sampel kontrol gen ACC1, dengan nilai Ct 36,69. Sedangkan nilai Ct pada tiga sampel lain tidak dapat diidentifikasi. Faktor kualitas (kemurnian dan konsentrasi) RNA, desain primer, prosedur perlakuan dan penyimpanan sampel dan RNA, serta perbedaan level dan modifikasi ekspresi gen diduga menyebabkan tidak teridentifikasinya nilai Ct pada ketiga sampel tersebut. Studi literatur dilakukan untuk mengetahui tren ekspresi kedua gen dibawah cekaman Nitrogen dan Silika. Gen ACC1 cenderung mengalami peningkatan ekspresi (upregulated), sedangkan ekspresi gen ACC2 dapat meningkat (upregulated) maupun menurun (downregulated).

Skeletonema costatum is a potential diatom microalgae which may be used as biodiesel raw material because of its high TAG lipid contents. TAG lipid can be induced by Nitrogen and Silica stress condition through the shifting of photosynthetic pathway to fatty acid biosynthesis pathway. Fatty acid biosynthesis were initiated by ACC1 and ACC2 gene. This research is conducted to know the expression of ACC1 and ACC2 genes under physiological stress conditions of Nitrogen (N) and Silica (Si) nutrients in Skeletonema costatum microalgae. The method used is gene expression analysis in samples of S. costatum RNA extract from a previous study by using qRT-PCR.
The results of gene expression analysis showed that there were 1 of 4 samples that could be identified by qRT-PCR, which is the control sample of ACC1 gene, with a 36.69 of Ct value. While the Ct values in the other three samples could not be identified. The quality factors (purity and concentration) of RNA, primer design, treatment and storage procedures for samples and RNA, also differences in levels and modification of gene expression allegedly caused the unidentified Ct values in the three samples. Literature study was conducted to determine the trend of two genes expression under Nitrogen and Silica stress. The ACC1 gene tends to increase its expression (upregulated), while the ACC2 gene expression may be increase (upregulated) or decrease (downregulated).
