Abstract

Keanekaragaman bakteri dapat ditinjau dari morfologi,
fisiologi, genetika maupun molekularnya. Keanekaragaman dapat dikaji melalui pendekatan taksonomi numerik fenetik. Beberapa anggota genus Bacillus berpotensi memiliki kemampuan resisten terhadap Fe sehingga dapat dijadikan sebagai agen bioremediasi. Penelitian ini bertujuan untuk mengetahui keanekaragaman Bacillus yang resisten Fe dari lahan Eco Urban Farming ITS dengan pendekatan taksonomi numerik fenetik. Metode yang digunakan meliputi isolasi bakteri, seleksi bakteri resisten logam Fe, karakterisasi morfologi, fisiologi, dan reaksi biokimia. Identifikasi sampai tingkat genus dan pengelompokan berdasarkan UPGMA (Unweighted Pair Group Method with Arithmetic Average). Isolat terseleksi sebanyak 7 isolat yang resisten logam
Fe sampai dengan 120 ppm. Pada generic assignment
menggunakan profile matching menunjukkan 5 isolat
teridentifikasi sebagai Bacillus, yaitu isolat UF1, UF4, UF5, UF6 dan UF8, sedangkan dua isolat bukan Bacillus yaitu UF3 dan UF7. Taksonomi numerik menggunakan konstruksi dendogram didapatkan 2 Cluster yaitu Cluster A dan Cluster B yang mempunyai indeks similaritas 64% dan menunjukkan bahwa
anggota Cluster A berbeda spesies dengan anggota Cluster B.

The diversity of bacteria can be viewed from the
morphology, physiology, genetics and molecular. Diversity can be studied through a phenetic numerical taxonomic approach. Some members of the genus Bacillus have the potential to be resistant to Fe so that they can be used as bioremediation agents. This study
aims to determine the diversity of Fe-resistant Bacillus from ITS Eco Urban Farming soil with a phenetic numerical taxonomic approach. The methods used include bacterial isolation, selection of Fe metal resistant bacteria, characterization of morphology, physiology, and biochemical reactions. Identification to genus level and grouping based on UPGMA (Unweighted Pair Group Method with Arithmetic Average). The selected isolates were 7 isolates that were resistant to Fe metal up to 120 ppm. In the generic
assignment using profile matching, 5 isolates were identified as Bacillus, namely UF1, UF4, UF5, UF6 and UF8, while two isolates were not Bacillus, namely UF3 and UF7. Numerical taxonomy using dendrogram construction obtained 2 clusters, namely Cluster A and Cluster B which have a similarity index of 64% and indicate that members of Cluster A are different species from members of Cluster B.
