Abstract

Nannochloropsis oculata merupakan salah satu jenis
mikroalga hijau yang hidup di laut. Kultur in vitro dengan
memberikan cekaman oksidatif pada mikroalga dilakukan untuk
menginduksi sel mikroalga menghasilkan senyawa bioaktif yang
bernilai ekonomis. Namun cekaman oksidatif akan mengakibatkan pembelahan sel terhambat dan perubahan morfologi, biokimia serta fisiologi sel mikroalga. Tujuan dari penelitian ini adalah untuk mengetahui morfologi berupa bentuk, warna, dan ukuran sel serta biomassa N. oculata pada cekaman NaCl 0 M, 0,3 M, 0,5 M, dan 0,7 M secara in vitro.
Rancangan penelitian menggunakan Rancangan Acak Lengkap (RAL) satu faktor yaitu variasi konsentrasi NaCl dengan
5 kali pengulangan. Parameter yang diamati adalah biomassa
kultur sel yang dianalisa secara kuantitatif dan morfologi sel meliputi bentuk, warna dan ukuran sel yang dianalisa secara deskriptif kualitatif. Hasil penelitian menunjukkan cekaman salinitas berpengaruh terhadap biomassa sel mikroalga N. oculata dimana biomassa perlakuan salinitas 0 M sebesar 9,197 mg/l berbeda signifikan dengan perlakuan salinitas 0.3 M sebesar 4,935 mg/l, perlakuan salinitas 0.5 M sebesar 4,865 mg/l, dan perlakuan salinitas 0.7M sebesar 3,022 mg/l. Pada pengamatan morfologi sel, perlakuan peningkatan salinitas menghasilkan bentuk sel lebih bulat (plump), berukuran lebih besar, dan warna sel berwarna hijau kemerahan.

Nannochloropsis oculata is a type of green microalgae that
lives in the sea. In vitro culture by applying oxidative stress to microalgae was carried out to induce microalgae cells to produce bioactive compounds of economic value. However, oxidative stress will result in inhibited cell division and changes in morphology, biochemistry and physiology of microalgae cells. The purpose of this study was to determine the morphology in the form of shape, color, and cell size as well as the biomass of N. oculata under NaCl stress of 0 M, 0.3 M, 0.5 M, and 0.7 M in vitro.
The research design used a completely randomized design
(RAL) with one factor, namely variations in NaCl concentration with 5 repetitions. Parameters observed were cell culture biomass which was analyzed quantitatively and cell morphology including shape, color and size of cells which were analyzed descriptively qualitatively. The results showed that salinity stress had an effect on the microalgae cell biomass of N. oculata where the biomass of 0 M salinity treatment was 9,197 mg/l significantly different from 0.3 M salinity treatment of 4,935 mg/l, 0.5 M salinity treatment of 4,865 mg/l, and salinity treatment. 0.7M at 3,022 mg/l. In the observation of cell morphology, treatment with increasing salinity resulted in a more rounded shape of cells (plump), larger in size, and reddish-green in color.
