Abstract

Pewarna reactive black 5 (RB5) banyak digunakan untuk pewarnaan tekstil dan ditemukan dalam air limbah. Pada penelitian ini dilakukan studi optimasi parameter yang meliputi konsentrasi pewarna RB5, waktu inkubasi, suhu, pH dan kondisi didiamkan serta digoyang terhadap kemampuan bakteri Ralstonia pickettii untuk Biodekolorisasi pewarna RB5 dalam media cair Luria Bertani broth (LB). Beberapa parameter seperti konsentrasi pewarna, masa inkubasi, suhu, pH, serta kondisi inkubasi diam atau digoyang digunakan dalam penelitian ini untuk menyelidiki pengaruhnya terhadap laju dekolorisasi RB5. Hasil penelitian menunjukkan bahwa R. picketti mampu mendekolorisasi 50 mg/L RB5 secara optimum dalam waktu 72 jam, suhu 40 °C dan pH 7 dan dalam kondisi didiamkan dengan persen dekolorisasi RB5 didapatkan hingga 92,79%, sedangkan dalam kondisi inkubasi digoyang dengan kecepatan 140 rpm dekolorisasi RB5 hanya mencapai 51,43%. Hasil menunjukkan bahwa R. pickettii dengan kondisi optimumnya memiliki kemampuan mendekolorisasi pewarna RB5.

Reactive black 5 dye (RB5) is widely used for dyeing textiles and found in wastewater. In this research, an optimization study of the parameters of RB5 dye including concentration, incubation time, temperature, and pH as well as static and shaked condition was conducted on the ability of the bacterium Ralstonia pickettii to biodegrade RB5 dye in Luria Bertani broth (LB) liquid media. Several parameter such as dye concentration, incubation time, temperature, pH, and static/shake condition ware used to in the present study to investigate their effect on the rate of RB5 decolorization. The results showed that R. pickettii was able to optimally degrade 50 mg/L RB5 within 72 hours, temperature 40 °C and pH 7 under static condition with the percentage of RB5 decolorization up to 92.79%, while in shaking condition at 140 rpm, RB5 decolorization was only 51.43%. The results showed that R. pickettii with its optimum conditions had the ability to decolorize RB5 dyes.
