Abstract

Gelatin merupakan emulsifier yang diperoleh dari hidrolisis parsial kolagen dan banyak digunakan untuk kebutuhan sahari-hari, lebih dari 60% total produksi gelatin digunakan oleh industri pangan, sekitar 20% industri fotografi, 10% oleh industri farmasi, dan 10% lainnya untuk industri kosmetik. Kebutuhan gelatin di Indonesia dipenuhi dengan cara impor dari Negara Perancis, Jerman, dan India. Berdasarkan data Kemenperin tahun 2012, impor gelatin tahun 2007-2011 meningkat 20,26% dengan nilai impor tahun 2011 mencapai 25.036,10 (ribu US dollar). Sehingga impor gelatin diperkirakan akan terus meningkat pada tahun-tahun berikutnya.
Pada penelitian ini penulis memanfaatkan limbah sisik ikan kakap untuk dijadikan gelatin dan mengetahui pengaruh konsentrasi pada produksi gelatin dari sisik ikan kakap. Proses pembuatan gelatin terdiri dari beberapa tahap. Tahap yang pertama adalah pre-treatment dengan memperkecil ukuran sisik ikan kakap. Tahap kedua adalah degreasing yaitu merendam sisik ikan kakap dalam air mendidih selama 30 menit. Tahap ketiga adalah demineralisasi dengan merendam sisik ikan kakap ke dalam larutan asam asetat dan asam sitrat dengan konsentrasi 3%, 4%, 5%, 6% dan 7% selama 3 hari sampai terbentuk ossein. Tahap keempat adalah menetralkan ossein dengan NaOH 1N sampai pH netral (pH 7). Tahap kelima adalah hidrolisis dengan memasukkan ossein yang sudah netral ke dalam labu leher tiga kemudian melakukan hidrolisis dengan waterbath pada suhu 70oC selama 3 jam disertai dengan pengadukan. Tahap selanjutnya adalah centrifuge, hasil hirolisis dicentrifuge dengan kecepatan 80 rpm selama 30 menit. Tahap yang terakhir adalah pengeringan, padatan hasil sentrifuge dimasukkan kedalam oven bersuhu 50oC selama 24 jam.
Pada percobaan yang dilakukan, didapatkan hasil gelatin yang paling optimal adalah gelatin yang diperoleh dari perendaman asam asetat 7% dengan rendemen sebesar 6,44%, nilai pH sebesar 6,66, kadar air sebesar 6%, kadar abu sebesar 2,91%. Hasil optimum juga didapat dari gelatin dengan perendaman asam sitrat 6% dengan rendemen sebesar 6,26%, nilai pH sebesar 6,35, kadar air sebesar 1%, kadar abu sebesar 1,98.

Gelatin is an emulsifier obtained from partial hydrolisis and is widely used for daily needs, over 60% of gelatin production is used for the food industry, about 20% is used for the photography industry, 10% is used for the pharmaceutical industry, and 10% is used for the cosmetics industry. The need for gelatin in Indonesia is filled with imports from France, Germany, and India. Based on data from the Ministry of Industry in 2012, the import of gelatin in 2007-2011 increased by 20.26% with import value in 2011 reaching 25,036.10 (thousand US dollars). So that gelatin import is expected to increase in the following years.
In this experiment, the authors utilize snapper scale’s waste to be used as gelatin and determine the effect of concentration on gelatin production from snapper scales. The process of making gelatin consists of several stages. The first stage is pre-treatment by minimizing the size of the snapper scales. The second stage is degreasing that soak the scales of snapper fish in boiling water for 30 minutes. The third stage is demineralization by soaking the snapper's scales into a solution of acetic acid and citric acid with concentrations of 3%, 4%, 5%, 6%, and 7% for 3 days until ossein is formed. The fourth stage is to neutralize ossein with 1 N NaOH until the pH is neutral (pH 7). The fifth stage is hydrolysis by inserting a neutral ossein into a three-neck flask and then hydrolyzing with a waterbath at 70°C for 3 hours accompanied by stirring. The next stage is the centrifuge, the result of hydrolisis is centrifuged with the speed of 80 rpm for 30 minutes. The last stage is drying, the solid of centrifuge is put into the oven at 50°C for 24 hours.
Based of the experiment, the most optimal gelatin result is gelatin obtained from 7% acetic acid immersion with yield of 6.44%, pH value of 6.66, water content of 6%, ash content of 2.91%. The optimum result also obtained from gelatin with 6% citric acid immersion with yield of 6.26%, pH value of 6.35, water content of 1%, ash content of 1.98.
