Abstract

Kolagen merupakan salah satu kelompok protein yang tidak larut air, yang keberadaannya mencapai 30% dari seluruh protein penyusun tubuh manusia. Peranan kolagen dalam tubuh manusia sebagai struktur organik pembangun tulang, gigi, sendi, otot dan kulit. Kolagen merupakan salah satu komponen yang terkandung di dalam kosmetik. Hal ini dikarenakan kolagen memiliki kemampuan untuk memberikan sifat elastis pada kulit dan dapat megurangi keriput yang terjadi sebagai efek dari penuaan. Manfaat lain kolagen adalah untuk membuat kulit tetap elastis dan sehat serta menghilangkan bekas luka pada kulit. Proses pembuatan kolagen dari tulang sapi terdapat 4 tahap. Tahap pertama merupakan tahap degreasing. Tahap degreasing merupakan tahap pembersihan tulang sapi dari kotoran selain tulang seperti lemak atau daging. Mencuci bersih tulang sapi, kemudian didiamkan selama 5-10 menit untuk menghilangkan kadar airnya, Setelah didiamkan, tulang dimasukkan di dalam tangki presto selama 3 jam untuk mengilangkan lemak yang melekat pada tulang. Selanjutnya tulang di demineralisasi untuk menghilangkan mineral (impurities) menggunakan solven asam yaitu asam asetat atau asam sitrat dengan konsentrasi 0,5 N; 0,75 N; 1 N; 1,25 N; dan 1,5 N selama 5 jam menggunakan stirrer pada suhu ruang. Setelah di demineralisasi tulang sapi dinetralkan dengan NaOH 10% air hingga pH netral dan dicuci bersih dengan aquades. Kemudian tahap berikutnya adalah ekstraksi dengan cara merendam tulang sapi ke dalam larutan asam asetat atau asam sitrat dengan ratio (1:5) pada konsentrasi 0,5 N; 0,75 N; 1 N; 1,25 N; dan 1,5 N selama 5 jam pada suhu operasi 40ºC serta menggunakan stirrer. Setelah itu, memisahkan antara ossein dan ekstraknya (larutan kolagen), kemudian melakukan penetralan dengan NaOH 10% hingga pH netral serta memisahkan larutan kolagen dengan residu. Setelah terpisahkan, memasukkan larutan kolagen ke dalam sentrifuge sehingga terbentuk dua lapisan antara kolagen basah dan solven. Kemudian hasil seluruh kolagen dikeringkan menggunakan oven dengan suhu 40º C selama 3 hari. Selanjutnya, tahap analisa yang meliputi analisa rendemen, pH, kadar air kolagen, kadar abu, organoleptik, FTIR, GC-MS, dan XRF. Didapatkan hasil yang paling optimal dengan pelarut asam asetat pada konsentrasi 1,25 N didapatkan rendemen sebesar 9,98% (w/w), kadar air sebesar 8,44% (w/v), kadar abu sebesar 0,74% (w/w), serta pH sebesar 6,66. Sedangkan pada solven asam sitrat pada konsentrasi 1,5 N dengan rendemen sebesar 10,65% (w/w), kadar air sebesar 7,95% (w/v), kadar abu sebesar 0,34% (w/w), serta pH sebesar 6,03.==========================================================================================Collagen is one of the groups of water-insoluble proteins, whose presence reaches 30% of all human body building proteins. The role of collagen in the human body as an organic structure builders bones, teeth, joints, muscles and skin. Collagen is one of the components contained in cosmetics. This is because collagen has the ability to provide elastic properties to the skin and can reduce wrinkles that occur as an effect of aging. Another benefit of collagen is to keep skin elastic and healthy and remove scars on the skin. There are 4 stages of making collagen from bovine bones. The first stage is the degreasing stage. The degreasing stage is the stage of cleansing cow bones from feces other than bones such as fat or meat. Wash off the cow bone, then stand for 5-10 minutes to remove the water content, After being silenced, the bone is inserted in the presto tank for 3 hours to remove the fat attached to the bone. Furthermore, bone is demineralized to remove impurities using acid solvents namely acetic acid or citric acid with a concentration of 0.5 N; 0.75 N; 1 N; 1.25 N; and 1.5 N for 5 hours using a stirrer at room temperature. After demineralization of beef bones neutralized with 10% NaOH solution to neutral pH and washed with distilled water. Then the next step is extraction by soaking the cow bone into a solution of acetic acid or citric acid with a ratio (1: 5) at a concentration of 0.5 N; 0.75 N; 1 N; 1.25 N; and 1.5 N for 5 hours at 40ºC operating temperature and using a stirrer. After that, separate the ossein and the extract (collagen solution), then neutralize with 10% NaOH solution to neutral pH and separate the collagen solution with residue. Once separated, insert the collagen solution into the centrifuge so as to form two layers between wet collagen and solvent. Then the whole collagen product was dried using oven at 40º C for 3 days. Furthermore, the analysis phase includes yield analysis, pH, collagen water content, ash content, organoleptic, FTIR, GC-MS, and XRF. The most optimal results obtained with acetic acid solvent at a concentration of 1.25 N obtained a yield of 9.98% (w / w), water content of 8.44% (w / v), ash content of 0.74% (w / w), and pH of 6.66. While in citric acid solvents at a concentration of 1.5 N with a yield of 10.65% (w / w), water content of 7.95% (w / v), ash content of 0.34% (w / w), and pH of 6.03.
