Abstract

Azotobacter memiliki kelebihan dibandingkan dengan
bakteri rhizosfer lainnya karena Azotobacter dapat menambat N
atmosferik. Azotobacter juga mampu mensintesis hormon
pertumbuhan IAA, golongan auksin alami yang paling melimpah
dan terlibat dalam regulasi pertumbuhan dan perkembangan
tanaman. Berdasarkan kemampuannya tersebut dilakukan
penelitian untuk menguji potensi Azotobacter yang diisolasi dari
Eco Urban Farming ITS sebagai penghasil IAA. Uji kualitatif
dilakukan untuk penapisan Azotobacter yang mempunyai indikasi
menghasilkan hormon IAA dengan metode Salkowksi pada media
NA triptofan. Selanjutnya dilakukan uji kuantitatif untuk
mengetahui konsentrasi hormon IAA yang mampu dihasilkan oleh
isolat Azotobacter dengan metode determinasi kolorimetri yang
ditunjukkan dengan pereaksi Salkowksi pada media Nutrient
Broth yang disuplementasi triptofan. IAA yang terbentuk
diukur menggunakan spektrofotometer dengan λ=530 nm. Isolat
yang mampu menghasilkan IAA adalah Azotobacter A1a, A1b,
A3, A6, A9, dan A10. Isolat Azotobacter A1a, A9 dan A10 dapat
memproduksi IAA lebih besar dibanding yang lain. Konsentrasi
IAA terbesar dihasilkan oleh isolat Azotobacter A10 mencapai
9,45 ppm, diikuti isolat Azotobacter A9 sebesar 7,38 ppm dan
isolat Azotobacter A1a sebesar 5,40 ppm.

Azotobacter has a beneficial value compared with other
rhizosphere bacteria because Azotobacter can fix atmospheric
nitrogen. Azotobacter is also able to synthesize growth hormone
IAA, the most abundant natural auxin group,and it is involved in
the regulation of plant growth and development. Based on that
potential, the research was conducted to test the potential
Azotobacter which was isolated from Eco Urban Farming ITS, as
a producer of IAA. Qualitative test was done to screen
Azotobacter that indicate producing hormones IAA with
Salkowksi method on NA tryptophan. Then, quantitative test was
done to measure the amount of the concentration of IAA hormone
which was produced by Azotobacter isolate using colorimetric
determination method, which was shown by Salkowksi reagent on
Nutrient Broth medium, that is supplemented by tryptophan.
Formed IAA was measured using spectrophotometer λ=530nm.
The result shown that the isolate that can produce IAA is
Azotobacter A1a, A1b, A3, A6, A9, and A10. Azotobacter A1a, A9
and A10 can produce IAA higher than others. The largest
concentration of IAA produced by isolate Azotobacter A10 which
can reach 9.45 ppm, followed by A9 isolate Azotobacter with 7.38
ppm and isolate Azotobacter A1a with 5.40 ppm.
