Abstract

Malachite Green (MG) merupakan salah satu pewarna dasar (basic dye) yang dapat digunakan dalam industri teksil. MG bersifat sifat teratogenik dan karsinogenik. MG sangat beracun bagi sel mamalia karena dapat menimbulkan tumor hati dan juga berbahaya bagi lingkungan. Penggunaan MG secara berlebih dapat menimbulkan bahaya bagi lingkungan dan manusia sehingga diperlukan cara untuk menguranginya. Penelitian ini mempelajari tentang dekolorisasi pewarna MG oleh kultur bakteri Bacillus subtilis. Variasi yang digunakan adalah konsentrasi, waktu inkubasi, suhu inkubasi, pH dan statis shaker. Hasil yang didapatkan menunjukkan bahwa MG dengan konsentrasi 50 mg/L dapat didekolorisasi pada kondisi optimumnya, dalam media Luria-Bertani Broth (LB), pH 7 dengan waktu inkubasi 36 jam pada suhu 40 °C. Kultur bakteri B. subtilis pada keadaan optimum mampu mendekolorisasi MG hingga mencapai 99,39%. Hasil menunjukkan bahwa B. subtilis dengan kondisi optimumnya memiliki kemampuan dalam mendegradasi pewarna MG.

Malachite Green (MG) is one of the basic dyes that can be used in the textile industry. MG is teratogenic and carcinogenic. MG is very toxic to mammalian cells because it can cause liver tumors and is also harmful to the environment. Excessive use of MG can pose a danger to the environment and humans, so it is necessary to find a way to reduce it. This study investigated the decolorization of MG dye by the bacterial culture of Bacillus subtilis. The optimum conditions were determined by variations of MG concentrations, incubation times, incubation temperatures, pH and static shaker. The results showed that MG with a concentration of 50 mg/L could be decolorized well at its optimum condition, in Luria-Bertani Broth (LB) medium, pH 7 with an incubation time of 36 hours at 40 °C. Bacterial culture of B. subtilis under optimum conditions was able to decolorize MG up to 99.39%. The results showed that B. subtilis with its optimum conditions had the ability to degrade MG dyes.
