Abstract

Indonesia merupakan salah satu produsen lada terbesar di dunia. Dalam pengolahan lada, sekitar 80% kulit dan tangkainya dibuang sebagai limbah biomassa. Limbah biomassa lada mengandung selulosa yang dapat digunakan sebagai prekursor untuk sintesis Karboksimetil Selulosa (KMS). KMS merupakan biopolimer turunan selulosa anionik yang larut dalam air. KMS digunakan untuk berbagai aplikasi di sejumlah industri termasuk makanan, perawatan pribadi, farmasi, ladang minyak dan industri kertas. KMS memiliki sifat unik seperti, viskositas yang tinggi, bersifat non-toksisitas, biokompatibilitas, biodegradabilitas, serta kemampuan pembentukan film yang baik dan biaya rendah. Pada penelitian ini dilakukan pengujian tentang aplikasi dari KMS dari limbah lada sebagai material penghantar obat terkontrol dengan model obat adalah propranolol (PPN). KMS diubah menjadi bentuk butiran dengan penambahan ion Fe3+ sehingga terbentuk ikatan silang. Hasil yang didapatkan dikarakterisasi dengan FTIR, XRD dan SEM. Setelah butiran KMS terbentuk dilakukan uji penggembungan dengan dua variasi pH yaitu pH 1,2 sebagai larutan model lambung dan pH 6,8 sebagai variasi larutan model usus. Didapatkan rasio penggembungan untuk butiran KMS pada pH 1,2 hingga mencapai 82,9% sedangkan untuk pH 6,8 mencapai 33,1%. Hal ini menandakan bahwa ikatan silang pada butiran KMS hampir sempurna (tinggi) sehingga nilai rasio penggembungan dari pH 1,2 lebih besar daripada pada pH 6,8. untuk hasil dari uji pemuatan obat yang berhasil temuat sebesar 1,7643 mg dengan nilai efisiensi adsorpsi sebesar 9,6%. Hasil dari uji pelepasan obat sesuai dengan kemampuan penggembungan yang telah diamati. Butiran KMS yang direndam pada pH 1,2 berhasil melepaskan obat hingga mencapai 22,8 mol/L sedangkan untuk pH 6,8 mencapai 1,1 mol/L.

Indonesia is one of the largest pepper producers in the world. In pepper processing, about 80% of the skin and stalks are disposed of as biomass waste. Pepper biomass waste contains cellulose which can be used as a precursor for the synthesis of Carboxymethyl Cellulose (KMS). KMS is a water-soluble anionic cellulose derivative biopolymer. Carboxymethyl Cellulose is used for a variety of applications in several industries including the food, personal care, pharmaceutical, oil field and paper industries. Carboxymethyl Cellulose has unique properties such as high viscosity, non-toxicity, biocompatibility, biodegradability, good film-forming ability, and low cost. In this study, a test was conducted on the application of carboxymethyl cellulose from pepper waste as a controlled drug delivery material with the drug model propranolol (PPN). Carboxymethyl cellulose is converted into beads form by the addition of Fe3+ ions so that cross-links are formed. The results obtained were characterized by FTIR, XRD and SEM. After the carboxymethyl cellulose beads were formed, a swelling test was carried out with two variations of pH, namely pH 1.2 as the gastric model solution and pH 6.8 as the intestinal model solution variation. The swelling ratio for carboxymethyl cellulose beads was obtained at pH 1.2 to reach 82.9% while for pH 6.8 it reached 33.1%. This indicates that the cross-linking of the carboxymethyl cellulose beads is almost perfect (high) so that the swelling ratio value of pH 1.2 is greater than at pH 6.8. for the results of the drug loading test which was successfully contained 1.7643 mg with an adsorption efficiency value of 9.6%. The results of the drug release assay agreed with the observed swellability. Carboxymethyl cellulose beads soaked at pH 1.2 managed to release the drug up to 22.8 mol/L while for pH 6.8 it reached 1.1 mol/L.
