Abstract

60% bagian Garcinia mangostana Linn. atau buah manggis teridentifikasi sebagai limbah, yang berasal dari kulit buah dan dibuang begitu saja. Beberapa penelitian menyatakan, bahwa kulit buah manggis memiliki kandungan senyawa bioaktif xanthone yang memiliki peran sebagai agen anti inflamasi, anti kanker, anti diabetes, anti bakteri, dan lain-lain, khususnya senyawa α-mangostin. Telah dilakukan penelitian dengan berbagai macam metode ekstraksi kulit buah manggis untuk mendapatkan yield ekstrak senyawa bioaktif terbaik. Tujuan penelitian ini yaitu mengetahui kondisi terbaik proses ekstraksi senyawa α-mangostin dari kulit buah manggis dengan metode Liquid-Liqud Extraction (LLE) serta mengetahui aktivitas antioksidannya. Pada penelitian ini, ekstraksi metode LLE dilakukan dengan menggunakan pelarut pilihan berbasis green solvent yang diharapkan memiliki selektivitas tinggi terhadap senyawa α-mangostin. Pengaruh polaritas pelarut, waktu ekstraksi dan suhu selama ekstraksi terhadap hasil ekstrak kulit buah manggis, dipelajari dalam penelitian ini. Analisis kualitatif dan kuantitatif kandungan α-mangostin pada ekstrak digunakan metode Kromatografi Lapis Tipis (KLT) dan Liquid Chromatography Mass Spectrometry-MS (LCMS-MS). Aktivitas antioksidan α-mangostin dianalisa menggunakan metode DPPH (1,1-diphenyl-2-picrihidazil) scavenging activity dan uji Total Phenolic Compound (TPC) dengan metode Folin-ciocalteu. Hasil penelitian menunjukkan antioksidan terbaik, yaitu ekstrak fraksi polar campuran pelarut Isopropil alkohol:Heksan:Air (1:1:1) pada kondisi operasi suhu 45°C dan waktu ekstraksi 1 jam dengan hasil %yield ekstrak 17,21; %inhibisi DPPH 65,90 (20 µg/mL); nilai TPC 1404,95 mg GAE/mg ekstrak (20 µg/mL); dan memiliki nilai IC50 pada 7,24 µg/mL. Sedangkkan %inhibisi DPPH larutan standart α-mangostin (20 µg/mL) bernilai 24,76.

60% of the Garcinia mangostana Linn. or mangosteen fruit is identified as waste, which comes from the skin of the fruit. It has been reported that the mangosteen peel’s contains bioactive compounds xanthones that plays as anti-inflammatory, anti-cancer, anti-diabetic, anti-bacterial agents etc., especially α-mangostin. Various research extraction methods have been conducted to get the best yield extracts of bioactive compounds. The aim of this study is to find out the best conditions of the extracting process α-mangostin from mangosteen peels’s with Liquid-Liqud Extraction (LLE) method and known its antioxidant activity. In this study, the LLE method was carried out using choice solvent (green solvent) which expected having high selectivity to α-mangostin. The effect of polarity solvent, extraction time and temperature during extraction on mangosteen peel extract were observed. Qualitative and quantitative analysis of the α-mangostin content in the extract used the Thin Layer Chromatography (TLC) method and Liquid Chromatography Mass Spectrometry-MS (LCMS-MS). Antioxidant activity of α-mangostin was analyzed using DPPH (1,1-diphenyl-2-picrihidazil) scavenging activity and Total Phenolic Compound (TPC) with the Folin-ciocalteu method. The results showed, the best antioxidants was at the polar fraction of extract from mixture solvent of Isopropyl alcohol:Hexane:Water (1:1:1) at operating conditions of 45 °C temperature and extraction time of 1 hour with %yield extract of 17.21; %DPPH inhibition 65.90 (20 µg/mL); TPC value 1404.95 mg GAE/mg extract (20 µg/mL); and IC50 value at 7.24 μg/mL. In comparison, %DPPH inhibition of standard solution α-mangostin (20 μg/mL) is 24.76.
