Abstract
Gelatin adalah produk alami yang diperoleh dari hidrolisis parsial kolagen. Sifat gelatin dipengaruhi oleh crosslink pada kolagen, di mana perbedaan crosslink dipengaruhi oleh profil asam amino dan gugus fungsi. Gelatin dari kulit ikan patin (GKIP) berpotensial sebagai bahan baku gelatin halal. Namun, GKIP memiliki kandungan prolin dan hidroksiprolin yang rendah sehingga ikatan silang yang terbentuk sedikit dan mengakibatkan film gelatin mudah lembek. Penambahan Transglutaminase (TGA) dapat mengkatalis ikatan silang antara glutamin dan lisin dalam protein sehingga sifat fisik gelatin lebih baik. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan TGA terhadap gugus fungsi dan profil asam amino pada gelatin kulit ikan patin. Kulit ikan patin diekstraksi dengan pre-treatment asam dan basa untuk memperoleh kolagen, kemudian didenaturasi menjadi gelatin, dan ditambahkan TGA 0,01, 0,05 gr dan tanpa TGA. Analisis gugus fungsi gelatin dilakukan dengan menggunakan FTIR (Fourier Transform Infrared) dan profil asam amino dilakukan dengan TLC (Thin Layer Chromatography). Hasil menunjukkan bahwa analisis gugus fungsi GKIP tanpa TGA, dengan penambahan TGA 0,01 dan 0,05 gr memiliki bilangan gelombang pada daerah serapan Amida A, I, II, dan III, namun bilangan gelombang pada daerah serapan Amida B hanya dimiliki oleh gelatin dengan penambahan TGA 0,05. Selain itu, GKIP TGA 0,01 memiliki lebih banyak puncak serapan dengan amplitudo yang tinggi. Pada ketiga sampel GKIP mengandung gugus fungsi O-H, N-H, C=O, C-N. Analisis profil asam amino menunjukkan bahwa asam amino yang muncul pada GKIP kontrol (18 asam amino), GKIP TGA 0,01 (8 asam amino), sedangkan pada GKIP TGA 0,05 profil asam amino tidak teramati pada uji TLC.

Gelatin is a natural product obtained from the partial hydrolysis of collagen. Gelatin properties are influenced by crosslinks in collagen, where the crosslinks differences are influenced by amino acid profiles and functional groups. Striped catfish skin gelatin (GKIP) has the potential as a raw material for halal gelatin. However, GKIP has a low content of proline and hydroxyproline so the cross-links formed are few and cause the gelatin film has a mushy texture. The addition of Transglutaminase (TGA) can catalyze cross-linking between glutamine and lysine so the physical properties of gelatin are better. This study aims to determine the effect of the addition of TGA on functional groups and amino acid profiles in striped catfish skin gelatin. Striped catfish skin was extracted with acid and base pre treatment to obtain collagen, then denatured into gelatin, and added TGA 0.01, 0.05 g and without TGA. The functional group analysis of gelatin was performed using FTIR (Fourier Transform Infrared) and the amino acid profile was performed using TLC (Thin-Layer Chromatography). The results showed that the analysis of the GKIP functional group without TGA, with the addition of TGA 0.01 and 0.05 gr have wave numbers in the Amide A, I, II, and III absorption regions, but the wave number in the Amide B absorption region was only in gelatin TGA 0.05. In addition, GKIP TGA 0.01 has more absorption peaks with high amplitude. The three GKIP samples contained functional groups O-H, N-H, C=O, C-N. Amino acid profile analysis showed that the GKIP control had 18 amino acids, GKIP TGA 0.01 (8 amino acids), while for GKIP TGA 0.05, no amino acid profile was observed in the TLC assay.
